    Objective To explore the effect of rTMS on the plasticity of the brain after ischemic injury. Methods SD rat MCAO-reperfusion model was established and then were divided into four groups. Different rTMS were applied within 7 days after the reperfusion. The neurological impairment evaluation score of rats were recorded during this period. Immunohistochemistry and Western blot techniques were used to detect GAP-43 and SYN protein expressions. Results rTMS could improve rehabilitation of neurofunction. rTMS could increase the protein expression levels of GAP-43 and SYN. The therapeutic effect of high frequency and high intensity of rTMS was statistically better than other groups (P<0.05). Conclusion The application of rTMS continuously could enhance the plasticity of the brain and improve neurofunction.

    Objective To explore the factors related to the recurrence of RVVC.Methods Samples were collected from the discharge of vagina,rectum,oral cavity in 55 patients and the discharge of balanus in 45 sexual partners of patients.Using SSCP analysis of internal transcribed spacer (ITS) region and sequence analysis of the 26S rDNA D1/D2 domain to identify the species of C. albicans of the samples. Genotypes of C. albicans strains were determined with use of SSCP combined to GeneScan analysis of C. albicans microsatellite locus I.Results The positive rates of vagina , oral and rectal Candida species colonization in the 55 women and 45 men were 100.0%(55/55)、61.8%(34/55)、1.8%(1/55) and 60.0%(27/45),respectively.And the percentage of Candida strains among the vagina , rectum and balanus were concurrent.In the same person,the coincidence of Candida albicans genotype between the discharge from vagina and rectum were 86.2%.Anymore,54.5% of the Candida albicans genotype of the discharge from the vagina of RVVC patients were same as the discharge from the balanus of her sexual partner.Conclusion The state of having candida on the rectum or the balanus of sexual partner could be one of the factors of RVVC.

    Objective The research aims to investigate the application of time series analysis method on time series datum, and establish forecasting model on cholecystitis incidence rate in Haixizhou region. Methods Based on the theory of time series model, through time series model some empirical studies into cholecystitis incidence rate have been made. ARMA and ARCH models are built to stimulate and forecast the pattern of the rate. Results The predicted result of ARCH model is more fitted than that of ARMA model and the ARCH model can describe the dynamic characteristics of cholecystitis incidence rate. Conclusion ARMA (4, 2)-ARCH (2) model can be used as the forecasting model of cholecystitis incidence rate in Haixizhou region.

    Objective To investigate the effects of GnRH analogue (GnRH-A) on secretion of LH and FSH, ovary histostructure and development in female rabbits, so as to provide the scientific bases for exploring further the mechanism of GnRH-A adjusting the reproductive functions. Methods Twenty-four female rabbits(Oryctolagus cuniculus)were divided randomly into experiment groups 1, 2, 3(EG-I, II, III)and control group (CG). Rabbit were subcutaneously injected with 1.0 mL GnRH-A antigen respectively at concentrations of 0.1g /L, 0.1g /L and 0.05g /L in EG-I, EG-II and EG-III. The rabbits in EG-II and EG-III were re-injected on 20 d. CG was as blank. Ten mL blood was collected on 0, 10, 20, 30, 40, 50, 60 and 70 d. The concentrations of FSH and LH in serum were assayed with ELISA. The ovary was collected aseptically on 70 d. The tissue sections were observed under the optical and electronic microscopes. Results Serum FSH and LH concentrations in EG-II and EG-III reached the peak on 40 d (20 d after re-injection). LH concentration in EG-II exceeded other 3 groups (P<0.05). Numbers of primary follicle increased. The ovarian cortex thickness(OCT)EG-II and EG-III thickened. The follicle vertical diameter(FVD) and follicle transverse diameter(FTD) enlarged, which was depedend upon the used dosage. GnRH-A could obviously speed up oocyte development by enlarging nucleus and chondriosome,increasing the quantities of mitochondrial cristae. The cortical granules and secretions in cytoplasm also raised. The zona pellucida and microvillus of the oocytes broadened and lengthened. Conclusion Injection of GnRH-A could enhance the synthesis and secretion of FSH and LH in female rabbits, and promote the development of ovary and follicle.

    Objective To explore the effects of Erlotinib combined with Celecoxib on apoptosis and expression of EGFR and COX-2 in lung cancer A549 cells.Methods A549 cells were observed under an inverted microscope ,MTT was used to measure the growth inhibitory effects of A549 cells by Erlotinib and Celecoxib.The cell apoptosis was studied byTUNEL and Hoechst33258 staining method . The cell cycle was detected by flow cytometer,and the expression of EGFR,COX-2 were determined by immunofluorescence. Results A great quantity of granules and vacuolus were observed in the combined treatment A549 ,with cell rounding and defluxion(P<0.05).The combination increased apoptosis of A549 (P<0.05) and significantly induced G1 phase arrest (P<0.05)and down-regulated expression of EGFR,COX-2 in A549 cells(P<0.05). Conclusions The results suggest that combination treatment with Erlotinib and Celecoxib showed significant synergistic inducing apoptosis and G1 phase arrest ,owing to down-regulating expressing both EGFR and COX-2.

    Objective To investigate role of Fosinopril (Fos) and Valsartan (Val) on klotho gene expression induced by Ang II and the relationship between klotho and TGF-β1. Motheds Culture cells, NRK-52E, were incubated with medias such as Ang II, Fos and Val. Five groups, control, AngII(10-7mol/L), AngII(10-7mol/L)+Fos(10-5mol/L), AngII(10-7mol/L)+Val(10-5mol/L) and AngII(10-7mol/L)+Fos(10-5mol/L)+Val (10-5mol/L) , were divided to observe the expression of the klotho and TGF-β1 mRNA and protein. The klotho and TGF-β1 mRNA were detected by reverse transcription – polymerase chain reaction (RT-PCR), immunohistochemistry (IHC) and Western Blotting (WB) was applied to determine the klotho and TGF-β1 protein expression. The linear relationship between klotho and TGF-β1 protein expression was analyzed by Pearson’s. Results 1. AngII induced an up-regulated expression of TGF-β1 mRNA , and associated with down-regulated klotho mRNA in NRK-52E as compared to control (all P<0.05); in cells incubated with Fos or Val or booth of all, the klotho mRNA expression was enheanced, and the TGF-β1 mRNA expression was diminished as compared to the AngII group(all P<0.05); combined Fos and Val could not enhance the expression of klotho gene, no significant different was showed as compared to the Fos group and Val group. 2. IHC and Western Blotting revealed that klotho protein was increased, TGF-β1 protein was decreased in Fos group, Val group and combination group as compared to AngII group, but no significantly different among the three groups was showed as compared to the Val group or Fos group. 3. Negative relationship was showed between klotho and TGF-β1 protein expression(r=-0.717, P<0.05). Conclusions Fosinopril and Valsartan can enhance the expression of klotho gene, and down-regulate TGF-β1 expression in NRK-52E, the increased level of klotho expression may be one of the important factor for preventing the renal fibrosis induced by TGF-β1.

    Objective To explore the effect of the combination of small dosage of VCR and Endostar on rhabdomyosarcoma cells and its mechanisms. Methods PLA-802 was cultured and treated with small dosage of VCR or Endostar singly or together. MTT was used to study the cells proliferation, and flow cytometry was for the cell cycle. While, RT-PCR and Western blot were for the expression of VEGF mRNA and protein levels. ELISA was for the generation of VEGF in supernatant. Results VCR (0.5 and 2.0μg/ml) or Endo (2.0 μg/ml) used singly inhibited the cell proliferation and promoted the apoptosis, and inhibited the expression and generation of VEGF. When the cells were treated with VCR and Endo together, the inhibitory effects of them were significantly strengthened. Conclusion The combination of VCR and Endostar could synergistically inhibit the expression and generation of VEGF and inducing the apoptosis in PLA-802 cells.

    Objects To study the relationship between the mitochondrial DNA (mtDNA) D-loop mutation and Huntington Disease. Methords (CAG)n repeat size and mtDNA D-loop fragment deletion or insertion were detected by PCR-DNA sequencing in two HD pedigree. Results normal individuals (CAG)n≤18 times, patient’s abnormal n≥40 times. There were no fragment deletion and insertion in the mitochondrial control region. Conclusions: Big fragment’s recombination in mtDNA control region maybe isn’t the important factor in HD pathogenic mechanism.

    Objective To investigate whether miRNA-1 may induce rat bone marrow mesenchymal stem cells (rMSCs) differentiation into cardiomyocyte-like cells. Methods rmiRNA-1 expression vector was constructed, rMSCs were isolated, amplified and identified. The fourth passage of rMSCs were transfected miRNA-1 plasmid with Lipofectamine2000 and miRNA-1 expression was then detected by quantitative real-time RT-PCR (qRT-PCR). The mRNA expression of cardiac-specific transcription factor GATA4, NKx2.5 and MEF2C were detected by RT-PCR,cTnI expression was measured by immunofluorescence,respectively,2,4,6 days. Results More than 90% of the cultured rMSCs were positive for CD29 and CD44, which are two significant markers of MSCs and negative for hematopoetic precursor cell markers CD34,leukocyte antigen CD45. After transfection of miRNA-1 in MSCs, the expression level of miRNA-1 was markedly enhanced. The mRNA expression of GATA4, NKx2.5 and MEF2C gradually increased from day 2 to day 6. The cTnI expression was detected by day 4,6. Conclusion miRNA-1 can induce MSCs differentiation into cardiomyocyte-like cells.

    Objective To explore the effects of hyperlipidemia in high-fat diet rat on expression of HSPA12B (heat shock protein A12B) in heart, arteriae aorta and liver tissue. Methods SD rats were fed a high-fat diet as model group (n=8, high-fat diet group) and were fed a normal diet as normal control group (n=8, normal diet group). Sixteen weeks later, serum lipid profiles including total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were detected. Heart, arteriae aorta and liver were stained with HE. HSPA12B mRNA expression in heart, arteriae aorta and liver were determined by RT-PCR, as well as protein expression by immunohistochemistry. Results Compared with the control group, high-fat diet group showed elevated TC (15.46±4.66)and TG (0.69±0.20)(both P<0.05). There were significant increase of HSPA12B mRNA in heart, arteriae aorta and liver tissues of model group than control group(P<0.01). Immunohistochemistry demonstrated: the positive expression of HSPA12B progressed, and the same result was analyzed by picture quantitative analysis techniques(P<0.05). Conclusion High-fat diet may lead to hyperlipidemia in rats, which may up-regulate HSPA12B expression in heart, arteriae aorta and liver tissues.

    Objective The relationship between the expression of HOGG1 in esophageal squamous cell carcinoma (ESCC) tissues and its 8-oxoG oxidative damage, its clinical and pathologic characteristic was investigated. Methods The expression of HOGG1 protein and the 8-oxoG oxidative damage between ESCC tissues and the adjacent tissues were compared by immunohistochemistry and western blot. And TUNEL kit was used to detect the apoptosis index (AI) of these tissues. The results showed that there were differences of HOGG1 expression among ECSS patients, and the different level of HOGG1 expression in ECSS tissues and the adjacent tissues. Results Decreased HOGG1 expression and increased 8-oxoG oxidative damage of ESCC tissues were found, compared with the adjacent tissues (P<0.05). Decreased HOGG1 expression of ESCC tissues was associated negatively with their 8-oxoG oxidative damage and AI, and associated with carcinoma vein encroachment and lymph node metastasis, but not associated with patients’ gender, age, pathological differentiation grade. Conclusion Postoperative routine detection of HOGG1 expression with the patients’ ESCC tissues may be useful for the guidance of making the patients’ chemotherapeutic schedule.

    Objective To observe the changes of mitochondria ultramicrostructure of mucinous cystadenocarcinoma(MC) and serous cystadenocarcinoma(SC) and to determine the stereological characteristic of the mitochondria of ovarian carcinoma cells and to investigate the value in clinical diagnosis of two cystadenocarcinoma Methods The ultramicrostructure changes of mitochondrion were observed with electronic microscope and the interrelation of volume density(Vvm), surface density(Sv,μm-1)and membrane surface density (Rsv, μm-1)of mitochondria of the cells in ovarian carcinoma were studied and compared with the normal control. Results Electron microscope reveals that the mitochondria swelling and cristae disorganizing were seen in ovarian carcinoma. The Vv and Sv of mitochondria of the MC cells were mitochondria increased obviously(0.891±0.04,P<0.05)。The Vv and Sv of mitochondria of the SC cells were increased markedly (0.267 ± 0.12 and 0.122 ± 0.06, P<0.01), while the its Rsv of mitochondria was declined obviously(0.584 ± 0.49,P<0.05)Conclusion There are obviously changes in the mitochondria of two types ovarian carcinoma.The number of mitochondria in former type is decreased, while the that of the latter is increased.
    Association of α-adducin G460T and AGT M235T gene polymorphisms with the risk of essential hypertension

    Objective To investigate the association between polymorphism of α-adducin G460T, Angiotensinogen (AGT) M235T genes and essential hypertension (EH) of Han nationality. Methods Mutagenically separated polymerase chain reaction (MS-PCR) and restriction fragment length polymorphism (RFLP-PCR) methods were used to detect the polymorphism of α-adducin G460T, AGT M235T genes in 305 EH patients and 305 age-matched, nationality normotensive controls (NT), biochemical index were also detected. Results The G460T polymorphism of α-adducin gene was no significantly association with EH, the genotype frequencies were 24.60%, 52.78% and 22.62% in the patient group and 28.53%, 48.85% and 22.62% in the control group, respectively. G and T allele frequencies were 50.98%, 49.02% in the patient group and 52.95%, 47.05% in the control group; The M235T polymorphism of AGT gene was significantly association with EH, the genotype frequencies were 18.69%, 52.46% and 8.85% in the patient group and 9.51%, 41.64% and 48.85% in the control group (P<0.01), respectively. M and T allele frequencies were 44.92%, 55.08% in the patient group and 30.33%, 69.67% in the control group(P<0.01); There were statistical significance of M235T genotype of AGT and G46OT genotype of α-adducin combined genes with EH, Every subset comparing had statistic significance in two groups(P<0.05), except GG genotype of α-adducin; Individuals carrying TT genotype of AGT M235T gene showed 2.35 high risk of EH (OR=2.35, 95%CI=1.69~3.28, P<0.01). Conclusions The TT genotype of AGT M235T gene may be related with increased risk of EH, and G460T polymorphism of α-adducin gene may have no relation with incidence of EH in Han nationality.

    Objective: This study is to investigate the effect of Rhein lysinate (RHL) on inducing human cervical cancer cell line HeLa apoptosis and the role of activation of c-Jun NH2-terminal kinase (JNK) in cell apoptosis. Methods: MTT assay was used to detect HeLa cell proliferation. Cell apoptosis was analyzed by flow cytometry. The protein associated with apoptosis and the protein phosphorylation of JNK were detected by Western blot. Results: RHL remarkably inhibited cervical cancer HeLa cells proliferation. The IC50 value for 48 h and 72 h treatment was 81.18 μmol/L and 72.38 μmol/L respectively. Apoptosis in HeLa cells was induced by RHL in a dose dependent manner. Mechanistically, RHL triggered HeLa cells apoptosis by increasing the levels of cleaved poly ADP-ribose polymerase (PARP) and caspase-3/7. In addition, activation c-Jun NH2-terminal kinase (JNK) was a critical mediator in RHL-induced growth inhibition. Inhibition of the phosphorylation of JNK by pharmacological inhibitors reversed the RHL-induced growth inhibition through decreasing the level of cleaved PARP and caspase-3/7. Conclusion: RHL suppresses HeLa cells growth and induce apoptosis through the activation of JNK-Caspase-PARP signal pathway, with the potential to be developed as a chemotherapeutic agent for cervical cancer.

    Objective To investigate the effect of transfection of hepatocyte cell adhesion molecule(HEPACAM) on apoptosis of renal cell carcinoma cells . Methods The vector containing HEPACAM gene was transfeceted into 786-0 cells, the expression of HEPACAM gene in 786-0 cells was confirmed by RT-PCR; The morphological changes of nucleus was observed by hoechst staining; The proliferation and apoptosis of 786-0 cells were analyzed by FCM; The expression of BAX、BCL-2、CYCLIN D1 were measured by western blotting. Results Up-regulated the expression of HEPACAM gene in 786-0 cells can arrest cell cycle at G0/G1 phase and induce cell apoptosis, and the expression of BAX was increased and the expression of BCL-2 and CYCLIN D1 was decreased. Conclusion These data indicated that ectogenic transfected HEPACAM gene may induce cell apoptosis and arrest cell cycle at G0/G1 phase in renal cell carcinoma cells.

    Objective To develop an effective and rapid method for screening recombinant hairpin RNA expression plasmids through reconstructing pSilencer3.1 -H1 vector. Methods The double-strand DNA fragement containing a NotⅠrestriction endonuclease site, the flanking sequences of which were not complementary, was annealed and ligated into small hairpin RNA (shRNA) expression vector pSilencer3.1-H1 to construct the pSilencer3.1-H1/NotⅠvector. With BamHⅠand Hind III, the pSilencer3.1-H1/NotⅠwas digested and ligated with siRNA expression cassette targeting JAK2 gene. The plasmid DNA of the positive clones was extracted and digested with NotⅠ. The un-digested vector was selected to identify by sequence analysis to construct the vector pSilencer3.1-H1/JAK2. After transfection of pSilencer3.1-H1/JAK2 into gastric cancer AGS cells, the JAK2 protein was detected by Western blotting. Results The sequencing results showed that pSilencer3.1-H1/NotⅠand pSilencer3.1-H1/JAK2 vectors were successfully constructed. The JAK2 protein was inhibited in transfected AGS cells. Conclusion The reconstructed pSilencer3.1-H1/ NotⅠvector could rapidly and efficiently screen shRNA expression vecteors.

    Objective To construct a targeting ribozyme (M1GS) which is specific to HCV genome. Methods According to the sequence of conservative sequence (5ˊUTR) of HCV genome, a guide sequence was designed and synthesized. And then by the PCR method, a kind of targeting ribozyme can be constructed by covalently linking the guide sequence to the 3ˊterminus of M1 RNA, the catalytic subunit of RNase P from Escherichia coli. Results The engineered ribozyme(M1GS-HCV/C67) is targeted to the 5ˊUTR of HCV genome, and can effectively cleave the substrate RNA segment in vitro. The cleavage is specific and the cleavage site is between 67nt and 68nt of the target region. Conclusion The M1GS-HCV/C67 we got here would be a useful experimental material to further study its cleavage activity in vivo, and can be even used for evaluating its anti-viral effect in the animal model. It was believed that this study would markedly facilitate the research of a general gene targeting agent for anti-HCV applications, and layed the foundation for developing a new nucleic acid drug of anti-HCV therapy.

    MicroRNAs (miRNAs) are a class of 20-24 nt non-coding RNAs that regulate gene expression primarily through posttranscriptional repression or mRNA degradation in a sequence-specific manner. It was reported that miRNAs have an important role on the regulation of new blood vessels. The thesis summarized miRNAs associated with therapeutic angiogenesis and reviewed latest studies of mechanism and clinic application of this novel therapy.

    Toll-like receptor (TLR) is a pathogen recognition receptor focused on by more people recently. It discriminates different pathogen-associated molecular patterns (PAMPs) and activates signaling cascades leading to immune responses. The specific TLR sign is conducted by adaptor proteins containing Toll–interleukin1 receptor (TIR) domains. The function is finished on MYD88 or no-MYD88/TRIF pathway, including five TIR adaptors: MYD88, MAL, TRIF, TRAM and SARM.

    Cachexia is the result of interaction between host and tumor,whose main manifestation are weight loss in short-term, malnutrition and so on.The inflammatory cytokines secreted by host and cachexia specific factor secreted by tumor are the main cause of cachexia . High-energy diet alone can not improve the condition. Recently, immunonutrition、traditional medicine and drug intervention can alleviate the weight loss and prolong survival in cachexia and thus has a promising prospect.
