Expression of HSPA12B in Heart, Aorta and Liver Tissue from Rats with Atherosclerosis

Abstract: Objective To explore the effects of hyperlipidemia in high-fat diet rat on expression of HSPA12B (heat shock protein A12B) in heart, arteriae aorta and liver tissue. Methods SD rats were fed a high-fat diet as model group (n=8, high-fat diet group) and were fed a normal diet as normal control group (n=8, normal diet group). Sixteen weeks later, serum lipid profiles including total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were detected. Heart, arteriae aorta and liver were stained with HE. HSPA12B mRNA expression in heart, arteriae aorta and liver were determined by RT-PCR, as well as protein expression by immunohistochemistry. Results Compared with the control group, high-fat diet group showed elevated TC (15.46±4.66)and TG (0.69±0.20)(both P<0.05). There were significant increase of HSPA12B mRNA in heart, arteriae aorta and liver tissues of model group than control group(P<0.01). Immunohistochemistry demonstrated: the positive expression of HSPA12B progressed, and the same result was analyzed by picture quantitative analysis techniques(P<0.05). Conclusion High-fat diet may lead to hyperlipidemia in rats, which may up-regulate HSPA12B expression in heart, arteriae aorta and liver tissues.
