HEPACAM induces apoptosis in renal cell carcinoma 786-0 cell line

Abstract: Objective To investigate the effect of transfection of hepatocyte cell adhesion molecule(HEPACAM) on apoptosis of renal cell carcinoma cells . Methods The vector containing HEPACAM gene was transfeceted into 786-0 cells, the expression of HEPACAM gene in 786-0 cells was confirmed by RT-PCR; The morphological changes of nucleus was observed by hoechst staining; The proliferation and apoptosis of 786-0 cells were analyzed by FCM; The expression of BAX、BCL-2、CYCLIN D1 were measured by western blotting. Results Up-regulated the expression of HEPACAM gene in 786-0 cells can arrest cell cycle at G0/G1 phase and induce cell apoptosis, and the expression of BAX was increased and the expression of BCL-2 and CYCLIN D1 was decreased. Conclusion These data indicated that ectogenic transfected HEPACAM gene may induce cell apoptosis and arrest cell cycle at G0/G1 phase in renal cell carcinoma cells.
