hsa-miR-4483促进人乳腺癌细胞系的增殖和迁移

hsa-miR-4483 promotes the proliferation and migration of breast cancer cell lines

摘要: 目的 探讨hsa-miR-4483在乳腺癌细胞系中表达及作用。方法 利用TargetScan找出hsa-miR-4483可作用c-Myc基因;利用RT-qPCR对hsa-miR-4483的表达量进行验证。转染MCF-7和MDA-MB-231,并设置NC阴性对照,Western blot检测hsa-miR-4483作用c-myc基因的表达量,利用MTT法和Transwell小室法验证乳腺癌细胞的增殖及迁移的能力。结果 hsa-miR-4483在MCF-7和MDA-MB-231中显著低表达(P<0.001,P<0.01);MCF-7和MDA-MB-231的c-myc蛋白表达量上调(P<0.05);hsa-miR-4483促进了MCF-7和MDA-MB-231细胞增殖和迁移(P<0.001)。结论 hsa-miR-4483可促进了乳腺癌细胞系的增殖以及迁移,其在乳腺癌的发展及其预后中起到调控的作用。

Abstract: Objective To explore the expression and function of hsa-miR-4483 in breast cancer cells. Methods Using TargetScan to find out how can-miR-4483 act on the c-Myc gene, and RT-qPCR verified the expression of hsa-miR-4483. Transfected MCF-7 and MDA-MB-231 and set up NC-negative controls, Western blot was used to detect the expression of the hsa-miR-4483 gene, MTT assay and Transwell chamber experiment were applied to verify the proliferation and migration of breast cancer cells. Results The expression of hsa-miR-4483 was significantly lower (P<0.001, P<0.01) in MCF-7 and MDA-MB-231(P<0.001, P<0.01) The up-regulation of c-myc in MCF-7 and MDA-MB-231 promoted cell proliferation and migration (P<0.001). Conclusions hsa-miR-4483 can promote the proliferation and migration of breast cancer cell lines, which plays a regulatory role in the development of breast cancer and its prognosis.
