敲除EZH2抑制人宫颈癌细胞系迁移和侵袭

Knockout of EZH2 inhibits migration and invasion of human cervical cancer cell lines

摘要: 目的 研究zeste基因同源蛋白2(EZH2)对人宫颈癌细胞系迁移和侵袭的影响,并初步探讨其作用机制。方法 利用CRISPR/Cas9敲除人宫颈癌HeLa和人子宫颈鳞癌SiHa细胞系中EZH2的表达。划痕实验检测细胞迁移;Transwell小室法检测细胞体外迁移和侵袭;实时荧光定量PCR 检测STAT3 mRNA 水平;Western blot 检测STAT3、p-STAT3蛋白表达。结果 CRISPR/Cas9能够敲除EZH2表达;敲除EZH2组中,HeLa和SiHa细胞的划痕愈合率24 h与48 h和Transwell小室迁移和侵袭穿膜细胞数目均低于对照组(P<0.05)。与对照组相比,敲除EZH2组的p-STAT3 蛋白水平明显降低(P<0.05)。结论 EZH2在宫颈癌中发挥促进宫颈癌细胞迁移和侵袭的作用,可能与靶向调控STAT3通路有关,具有作为宫颈癌治疗新靶点的潜能。

Abstract: Objective To explore the effect of EZH2 on the migration and invasion of human cervical cancer cell lines and its mechanism. Methods CRISPR/Cas9 was used to knockout EZH2 expression in HeLa and SiHa cell lines. The migration was detected by scratch test, and the migration and invasion in vitro was detected by Transwell migration and invasion test. The STAT3 mRNA level was detected by real-time fluorescence quantitative PCR, and the expression of STAT3 and p-STAT3 proteins was detected by Western blot. Results CRISPR/Cas9 could knockout EZH2 expression. The wound healing rate of HeLa and SiHa cells in EZH2 knockout group at 24 h and 48 h, and the counting of transmembrane cell migration and invasion in Transwell chamber were lower than those in control group(P<0.05). Compared with the control group, the protein level of p-STAT3 in EZH2 knockout group was significantly decreased (P<0.05). Conclusions EZH2 plays an important role in promoting the migration and invasion of cervical cancer cells, which may be related to the targeted regulation of STAT3 pathway, and is a potential target in the treatment of cervical cancer.
