阿尔茨海默病和糖尿病的关联:揭示一种新的治疗策略

Connecting Alzheimer's disease to diabetes: Underlying mechanisms and potential therapeutic targets

摘要: 阿尔茨海默病(AD)是2型糖尿病(T2D)的危险因素,反之亦然。越来越多的证据表明这些疾病在流行病学、临床和分子水平上都有关联。最近的研究揭示了AD和2型糖尿病具有共同的致病机制。在AD的动物模型中发现神经元胰岛素信号传导受损和内质网(ER)应激,类似于在T2D外周组织中的观察结果。这些研究揭示了一些新的糖尿病相关的机制,这些机制能够引起AD脑功能障碍。在这里,我们回顾了这两种疾病之间共同出现的机制的文献,并讨论如何从这些机制中探索在AD新的可能治疗靶点。

Abstract: Alzheimer's disease (AD) is a risk factor for type 2 diabetes(T2D) and vice versa, and a growing body of evidence indicates that these diseases are connected both at epidemiological, clinical and molecular levels. Recent studies have begun to reveal common pathogenic mechanisms shared by AD and type 2 diabetes. Impaired neuronal insulin signaling and endoplasmic reticulum (ER) stress are present in animal models of AD, similar to observations in peripheral tissue in T2D. These findings shed light into mechanisms leading to brain dysfunction of AD in T2D patients. Here, we review the literatures on selected mechanisms shared between these diseases and discuss how the identification of such mechanisms may lead to novel therapeutic targets in AD.
