Expression and effect of lncRNA GASL1 in breast cancer tissues and cell line

摘要: 目的 探讨lncRNA GASL1在乳腺癌中的表达及其对乳腺癌癌细胞增殖和凋亡的影响。方法 RT-qPCR检测癌组织、癌旁组织、乳腺癌细胞系(MDA-MB-231、MDA-MB-468和SK-BR-3)及正常乳腺细胞HBL-100中lncRNA GASL1的表达水平。MCF-7细胞经pcDNA3.1质粒过表达lncRNA GASL1,或siRNA干扰lncRNA GASL1表达后,运用MTT法检测细胞增殖;caspase-3活力检测试剂盒检测caspase-3活力;Western-blot 检测Bax和Bcl-2蛋白表达。结果 LncRNA GASL1在乳腺癌组织和乳腺癌细胞中表达水平降低(P<0.05);过表达LncRNA GASL1可抑制MCF-7的增殖,促进其凋亡(P<0.05);干扰LncRNA GASL1表达可促进MCF-7的增殖,抑制其凋亡(P<0.05)。结论 lncRNA GASL1在乳腺癌中表达水平降低,通过调节乳腺癌细胞增殖和凋亡参与乳腺癌的发生发展。

Abstract: Objective To investigate the expression of lncRNA GASL1 in breast cancer and the effect of lncRNA GASL1 on proliferation and apoptosis of breast cancer. Methods RT-qPCR was used to measure the expression of lncRNA GASL1 in breast cancer tissue and cells. pcDNA3.1 plasmid and siRNA were used to over-express and knockdown the expression of lncRNA GASL1 respectively in MCF-7 cells, and then measured the proliferation through MTT method, caspase 3 Activity Assay Kit was used to measure the caspase 3 activity. Moreover, the protein levels of Bax and Bcl-2 were tested by Western-blot. Results Lower lncRNA GASL1 levels in breast cancer tissue and cells compared with paracancerous normal tissues and normal breast cell HBL-100, respectively (P<0.05). Overexpression of lncRNA GASL1 inhibited MCF-7 proliferation and promoted apoptosis (P<0.05), while knockdown of lncRNA GASL1 increased proliferation and suppressed apoptosis (P<0.05). Conclusions The expression of lncRNA GASL1 is lower in breast cancer, which is involved in development of breast cancer through the regulation on breast cancer cell proliferation and apoptosis.
