转化生长因子β1促进大鼠子宫内膜纤维化

Transforming growth factor-beta 1 promotes endometrial fibrosis in rats

摘要: 目的 探讨大鼠子宫内膜纤维化中转化生长因子β1(TGF-β1)的表达及相关机制。方法 将SD 雌性大鼠随机分成假手术组(n=12)和模型组(n=12)。术后7、14及28 d收集双侧子宫组织,HE染色观察子宫内膜形态,动态观察子宫内膜病理改变和子宫内膜腺体数目;Masson 染色检测子宫内膜纤维化程度;免疫组织化学法检测TGF-β1及α-SMA 蛋白表达。结果 相比假手术组,模型组大鼠子宫结构破坏;术后7、14和28 d,子宫内膜TGF-β1蛋白表达显著升高(P<0.05);术后14和28 d,子宫内膜腺体数目明显减少、子宫内膜纤维化面积比例明显增加,α-SMA蛋白表达上调(P<0.05)。结论 TGF-β1可能通过激活肌成纤维细胞及相关信号通路进一步促进子宫内膜纤维化。

Abstract: Objective To investigate the expression of transforming growth factor-beta 1 in endometrial fibrosis in rats and related mechanisms. Methods Sprague-Dawley female rats were randomly divided into sham operation group (n=12) and model group (n=12). Uterine tissues were collected on days 7, 14 and 18 after surgery. HE staining and Masson staining were measured the number of endometrial glands and degree of endometrial fibrosis. The protein expression of TGF-β1 and α-SMA were measured by immunohistochemistry. Results Compared with the sham operation group, the uterine structure of the rats in the model group was significantly disrupted; there was a significant increase in TGF-β1 expression in the model group at 7 days,14 days, and 28 days of modeling; the number of endometrial glands decreased, the fibrotic area of endometrium increased and the expression of α-SMA was significantly higher at 14 days and 28 days of modeling. Conclusions TGF-β1 may promote endometrial fibrosis by activating myofibroblasts.
