尼可地尔预处理改善糖尿病合并冠心病模型大鼠心脏功能

Nicodil preconditioning improves cardiac function in rat models of diabeties complicated with coronary heart disease

摘要: 目的 探讨尼可地尔(Nic)对糖尿病合并冠心病(DM/CHD)大鼠心脏功能的影响及机制。方法 将大鼠分为:对照组、DM/CHD组、DM/CHD+Nic组和DM/CHD+Nic+shTRPV1组。超声心电图检测心脏功能参数。收集血液进行生化指标检测。HE染色、Masson三色染色和TUNEL染色分别检测心脏组织病理学改变、胶原沉积和细胞凋亡。Western blot检测蛋白质表达。结果 相较对照组,DM/CHD组心肌细胞直径和胶原堆积增加(P<0.05);凋亡细胞数及Bax和cleaved caspase-3表达增加(P<0.05),Bcl-2、TRPV1、p-PI3K和p-Akt表达降低(P<0.05)。相较DM/CHD组,DM/CHD+Nic组心肌细胞直径和胶原堆积减少(P<0.05);凋亡细胞数及Bax和cleaved caspase-3表达降低(P<0.05),Bcl-2、TRPV1、p-PI3K、p-Akt表达增加(P<0.05)。相较DM/CHD+Nic组,DM/CHD+Nic+shTRPV1组心肌细胞直径和胶原堆积增多(P<0.05);凋亡细胞数及Bax和cleaved caspase-3表达增多(P<0.05),Bcl-2、TRPV1、p-PI3K和p-Akt表达降低(P<0.05)。结论 Nic可能通过调节TRPV1-PI3K/Akt信号通路改善DM/CHD大鼠心脏功能。

Abstract: Objective To identify the impacts of nicorandil (Nic) on cardiac function of rats with diabetes mellitus and coronary heart disease (DM/CHD) and to explore the potential mechanism. Methods Rats were divided into: control group, DM/CHD group, DM/CHD+Nic group and DM/CHD+Nic+shTRPV1 group. The cardiac function parameters were detected by electrocardiogram. Blood was collected for biochemical index detection. Cardiac histo-pathological changes, collagen deposition and apoptosis were determined by HE, Masson trichrome and TUNEL staining microcopy respectively. The protein expression was analyzed by Western blot. Results In comparation to control group, the cardiomyocyte diameter and collagen accumulation were increased (P<0.05). Apoptotic cells and the expression of Bax and cleaved caspase-3 were increased (P<0.05),expression of Bcl-2, TRPV1, p-PI3K and p-Akt was decreased. Compared with DM/CHD group, myocardial cell diameter and collagen accumulation were reduced (P<0.05). Apoptotic cells and the level of Bax and cleaved caspase-3 were reduced (P<0.05).The expression of Bcl-2, TRPV1, p-PI3K and p-Akt was enhanced (P<0.05). Compared with DM/CHD+Nic group, myocardial cell diameter and collagen accumulation were increased (P<0.05). Apoptotic cells and the expression of Bax and cleaved caspase-3 were up-regulated (P<0.05).The expression of Bcl-2, TRPV1, p-PI3K and p-Akt was down-regulated. Conclusions Nic improves cardiac function in DM/CHD rats with potential mechanism of regulating TRPV1-PI3K/Akt signaling pathway.
