Objective To study the effects of G48A and alendronate in activation-depression-free-repeat (ADFR) scheme on ovariectomized rats.Methods Seventy-five three month old female Sprague-Dawley rats were randomly divided into 7 groups: sham-operated(S) group , ovariectomized (OVX)group, alendronate group(ALEN), G48A and alendronate group(non-ADFR group), and the other three groups were treated by the ADFR scheme with G48A and alendronate given intermittently by different time schedule :5-5-5d(group G48A1),10-10-5d(group G48A2), 14-4-7d(group G48A3). All rats were sacrificed 13 weeks after the operation.The BMD of femora and lumbar vertebra and the biomechanical parameters were measured.Results Body weight increased and BMD decreased in OVX compared those with S group.The lumb and vertebral BMD ,the vertebral elastic load ,the femoral flexural modulus and flexural rigidy coefficient in groups G48A1,2,3 and non ADFR group were significantly increased compared with those in group OVX.The BMDs of ADFR groups and non-ADFR group did not have significant difference compared with S group. Conclusion The bone loss decreased and the biomechanics improved in ovariectomized rats with the treatment of G48A and alendronate given by ADFR scheme.

Objective To investigate the effects of phytoestrogen α-zearalanol (α-ZAL) on vascular function in ovariectomized hyperhomocysteinemic rats. Methods Hyperhomocysteinemia was induced by fed with high-methionine diet. 40 adult female Wistar rats were assigned randomly into 5 groups: (1) Control (Con); (2) 2.5%Methionine (Met); (3) Ovariectomy+2.5%Methionine (OVX+Met); (4) Ovariectomy+2.5%Methionine+α-ZAL (OVX+Met+α-ZAL); (5) Ovariectomy+2.5% Methionine + 17β- estradiol (OVX+Met+17β-E2). 12 weeks later, blood was collected to analyze plasma estradiol and Hcy levels. Thoracic aortas were isolated to detect the response to phenylephrine (PE), acetylcholine (ACh) and sodium nitroprusside (SNP). Results The plasma Hcy levels were elevated in all rats fed with 2.5% methionine diets, especially in the OVX+Met group. In a dose-dependent manner, PE elicited significantly greater contraction in thoracic aorta rings taken from rats of OVX+Met than that taken from other groups and ACh elicited significantly less percentage relaxation in thoracic aorta rings. There was no significant difference in response to SNP between groups. Conclusion α-ZAL could remarkably improve vascular function in ovariectomized HHcy rats via attenuating elevation of plasma Hcy and endothelial cells damage, which is similar to 17β-E2.

Objective: to understand th population genetic characters of Mulam's head face. Methods: Nine genetics indexes,including mongoloid fold,eyefold of the upper eyelid,front tooth type,nasal profile,nostril form,chin projection,lobe type,hair point of the forehead and hair form were investigated for 309 middle schol students(120 males and 189 females)in Mulam nationality by using cluster random sampling method. Results: (1)The rate of Mongolia fold is 72.17% ,The rate of eyefold of the upper eyelid is 84.12%,The rate of shovel-shaped front tooth type is 96.12%,The rate of umbonate nasal profile is 6.47%,The rate of wide nostril is 79.29%,The rate of protruding chin is 29.13%,The rates of ear lobe is 74.11%,The rate of pointed hair point on forehead is 24.87% and The rate of curly hair form is 0.97% in Mulam nationality;(2)eight indexes showed no sexual difference except for The rate of eyefold of the upper eyelid;(3)These nine indexes showed extremly little correlations;Conclusions: There are differences between Mulam nationality and other nationalities in Population genetic characters of head face.

Objective To examine the effect of VEGF165 on expressions of JAGGED-1and NOTCH1 and the relationship between these expressions and epithelial- mesenchymal transition (EMT) induced by TGF-β1. Methods Cultured HK2 cells were incubated with TGF-β1(5μg/L) in the absence (group T) or presence (group T+V) of VEGF165 (100μg/L),respectively. Expressions of α-SMA and JAGGED-1mRNA were assessed with RT-PCR and the protein expressions were assessed with Western Blot at different time points,respectively. In the other experiment, HK2 cells were divided into four groups: negative control, incubated with TGF-β1(5μg/L) along, with VEGF165 (100μg/L) along, or with TGF-β1(5μg/L) and VEGF165 at different concentrations (0.1, 1, 10, 100μg/L) for 24 hours. Confocal microscope was used to detect expressions of α-SMA and E-cadherin. Protein and mRNA expressions of α-SMA, JAGGED-1and NOTCH1 were assessed with RT-PCR and Western Blot, respectively. Results Protein expression of both α-SMA and JAGGED-1decreased significantly in group T+V at 24h and 48h when compared to group T (P<0.05). VEGF165 reversed TGF β1-induced inhibition of E-cadherin and promotion of α-SMA, JAGGED-1and NOTCH1 significantly (P<0.05). Conclusions The results of the present study indicated that VEGF165 may partially inhibits TGFβ1-induced EMT of HK2 cells in vitro, which is probably contributed to the repression of JAGGED-1and NOTCH1 expressions. Further study is still needed.

Objective To study the effect of AdoHcy on proliferation and apoptosis of SVAREC and the content of VCAM-1 in the supernatant of SVAREC culture.Methods After SVAREC were treated with different concentrations of 3-deazaadenosine (DZA) for 24, 48 and 72h, the cellular AdoHcy level and the homocysteine (Hcy) were measured with different methods The cell growth and proliferation were determined by WST-1 assay. The percentage was analyzed with TUNEL methods. The content of VCAM-1 in the supernatant of SVAREC culture was analyzed with ELISA methods Results After SVAREC were treated with different concentrations of DZA, the content of Hcy decreased while the intracellular AdoHcy level increased.Compared with the control group, AdoHcy could inhibit the proliferation and induce apoptosis of SVAREC in a dose-dependent way and induce secretion of VCAM-1. Conclusions AdoHcy could inhibit the proliferation and induce apoptosis of SVAREC, induce secretion of VCAM-1.which may be one of mechanisms responsible for injury of aorta endothelial cell lead to atherosclerosis.

Objective To observe the phenomenon of epithelial-mesenchymal transition(EMT) and expression of P-glucoprotein(P-GP) in MCF-7 cells, induced by SNAIL, and reveal the effection of EMT enhance drug-resistance capacity of MCR-7 cells. Methods Constructed eukaryotic expression vector of SNAIL; and then transfected into MCF-7 cells. Expression of epithelial marker E-cadherin and interstitial marker Vimentin detected by immunofluorescence; drug resistance to adriamycin(ADM) was measured by MTT; expression of P-GP and SNAIL was detected by flow cytometry; SNAIL, E-cadherin, Vimentin and MDR1 mRNA were evaluated by Real-Time-PCR. Results Compared with parental MCF-7 cells, expression of E-cadherin decreased obviously, while Vimentin get up(P<0.01); drug resistance to ADM and expression of SNAIL, P-GP increased too(P<0.01), consistent with results of Real-Time-PCR. Conclusions EMT, induced by SNAIL, can enhance the drug-resistance capacity to ADM, mediated by P-GP, in breast cancer cell.

Objective To identify the pathogenic mutation in a large Chinese family with Charcot-Marie-Tooth disease (CMT). Methods Medical history of the proband and a detailed family history were collected. Necessary physical examination and nerve conduction velocity/EMG testing were performed to establish the diagnosis of CMT and a primary classification. Genomic DNA was extracted from peripheral blood samples of 16 family members. Five genetic loci for six CMT1 subtypes were screened by linkage analysis using microsatellite markers. Quantitative PCR was used to detect the genomic duplication encompassing the peripheral myelin protein 22 gene (PMP22). Results The CMT phenotype was transmitted in this family in an autosomal dominant pattern. Slow nerve conduction velocity (10-30m/s) was observed in all of the four patients who participated in the electrophysiologic testing. By linkage analysis, all CMT1 subtypes were excluded except the CMT1A and CMT1E locus, both of them involving the PMP22 gene on chromosome 17p11.2 region. Heterogeneous duplication of the PMP22 gene was detected in patients, but not in unaffected family members. Conclusion The disease in the family was ascertained to be CMT1A, and was caused by the PMP22 duplication.

Objective To study Adenosine inhibits lipopolysaccharide-induced human umbilical vein endothelial cell (HUVEC) apoptosis. Methods The cultured HUVEC was divided into 6 groups: Control group, LPS group, dimethylsulfoxide (DMSO) group, Low, Medium and High-dose adenosine group. the cell's shape structure were observed continuously by fluorescence microscope. TNF-α expression in the supernate was determined by ELISA. Cells's apoptosis was examined by Flow Cytometry. Results The TNF-α expression and cell apoptosis were significantly increased (p<0.05); Groups of different doses of adenosine can inhibit the expression of TNF-α and cell apoptosis (p<0.05). Conclusion Certain dose of adenosine can inhibits lipopolysaccharide-induced the HUVEC release inflammation medium, adenosine can also suppress cell apoptosis.

Objective To investigate the effect of nitric oxide (NO) on the expression of long type of leptin receptor (OB-Rb) in dorsal root ganglion (DRG) neurons cultured in vitro. Methods Sodium nitroprusside (SNP; 0, 0.01, 0.1 and 0.5 mmol/L) were used to treat the DRG neurons cultured in vitro. RT-PCR and Western Blot were used to detect the expression of long types of leptin receptor (OB-Rb) in DRG neurons. Results 0.1~0.5 mmol/L SNP can increase the expression of OB-Rb mRNA and protein Conclusion The modulation of NO on DRG neurons may be relative with the action of leptin.

Objective To investigate the effect of melatonin post-intervention on the expression of thioredoxin-1(TRX-1) in rats with acute necrotizing pancreatitis(ANP). Methods Male Spraque-Dawley rats were randomly allocated to control group, ANP group,melatonin post-intervention group with 24 rats in each group .The rats in ANP group received three intraperitoneal injections of 25 ml/kg body weight 6%L-arginine at an interval of 1 h to induce ANP. The rats in melatonin post-intervention group received intraperitoneal injections of 20ml/kg body weight 0.25% melatonin after ANP induction. Rats in control group received intraperitoneal injections of same amount of saline. Rats were sacrificed at 6, 12 and 24 h after ANP induction. The pathological evaluation of pancreatic tissues was performed. The concentrations of malondialdehyde and myeloperoxidase in pancreatic tissues were measured. The serum level of TRX-1 was measured by ELISA. The expressions of thioredoxin-1 and thioredoxin-1mRNA in pancreatic tissues were confirmed by immunohistochemistry and RT-PCR, respectively.Results Comparerd to the control group, pathological scores, malondialdehyde ,and myeloperoxidase of pancreatic tissues in ANP group were significantly increased(p < 0.05),while they in melatonin post-intervention group were significantly lower (p < 0.05) than those in ANP group. Comparerd to the control group,serum TRX-1 contents in ANP group significantly decreased(p < 0.05) at 6- and 12 h points, while they in melatonin post-intervention group were significantly higher (p < 0.05) than those in ANP group. the peak value of TRX-1 protein and TRX-1mRNA expressions in pancreatic tissues shifted from 12 h after ANP induction in ANP group to 6 h after ANP group in melatonin post-intervention group.Conclusions Melatonin post-intervention reduced the injury of pancreatic tissue may be through increased tissue and serum TRX-1 levels in ANP rats.

Objective To investigate the roles of angiopoietin-1(Ang-1) and endothelial nitric oxide synthase(eNOS) in pro-angiogenic effect of simvastatin after experimental myocardial infarction(MI). Methods 60 healthy adult SD rats were randomly divided into the sham operated group、control group、simvastatin group、simvastatin plus L-NAME(inhibitor of NOS)group and simvastatin plus AMG386(inhibitor of Ang-1)group; Left anterior descending coronary underwent permanent occlusion to establish the MI model. Rats with MI were administered simvastatin (1 mg/(kg·d) )、simvastatin plus L-NAME (40 mg/(kg·d) )、and simvastatin plus AMG386(10 mg/(kg·wk) ) respectively for 2 weeks. New microvessels in the ischemic area near the infarction myocardium were stained by CD31 and the density of new microvessels was dedected; Ang-1、eNOS and phosphoralated endothelial nitric oxide synthase at Ser1177 (p-eNOS)were evaluated by western blotting and RT-PCR assay. Results (1) simvastatin significantly increased the density of new microvessels(P<0.05),but L-NAME and AMG386 siglificantly inhibited the proangiogenic effect of simvastatin (P<0.05).(2) simvastatin significantly improved The expression of Ang-1、eNOS and p-eNOS (P<0.05), and AMG386 significantly decreased simvastatin induced upregulation of p-eNOS. Conclusion The proangiogenic effect of simvastatin is associated with increased expression of Ang-1、eNOS and p-eNOS, and phosphoralation of eNOS maybe the downstream pathway for Ang-1 induced angiogenesis.

Objective This study was to investigate the effect of nitric oxide (NO) on cell proliferation in gastric cancer cell line SGC-7901, and to investigate the relevant mechanisms. Methods MTT assay was used to detect the inhibitory effect of NO on cell growth. RT-PCR was used to detect the mRNA expression of C-FOS and C-JUN. Western blot was applied to study the protein expression of C-FOS and C-JUN. Results SNP inhibits the proliferation of SGC-7901 cells (P<0.05). SNP dose- and time-dependently decreased the expression of C-FOS, C-JUN mRNA and protein. Conclusions NO inhibits the proliferation of SGC-7901 cells, which is probably through NO down-regulated C-FOS and C-JUN expression.

Objective To study the effect of atorvastatin on the interaction of endothelial cells and mesangial cells exposed to high glucose and lysophosphatidylcholine( LPC ). Methods The model of intercellular interaction between human endothelial cells and human renal glomerular mesangial cells was established and divided into 5 groups: control, mannitol, high glucose and LPC, BN52021, atorvastatin. The level of fibronectin and PAF were determined by enzyme linked immunosorbent assay in the culture media. The expression level of platelet activating factor receptor(PAF-R)mRNA in mesangial cells was determined by real-time fluorescence quantitative PCR. Results 1.The level of fibronectin and PAF of high glucose and LPC group was higher than that of control group in co-culture and the monolayer cell culture (P<0.05 ). Intervented by high glucose and LPC, the level of fibronectin and PAF of co-culture was higher than that of the monolayer cell culture (P<0.05). The level of fibronectin of BN52021 or atorvastatin group was lower than that of high glucose and LPC group(P<0.05). The level of PAF of atorvastatin group was lower than that of high glucose and LPC group. 2. The expression level of PAF-R in mesangial cells of high glucose and LPC group was higher than that of control group in co-culture and the monolayer cell culture (P<0.05 ). Intervented by atorvastatin, the expression level of PAF-R in mesangial cells was lower than that of high glucose and LPC. Conclusions (1) Exposed to high glucose and LPC, endothelial cells and mesangial cells can interact with each other. (2) Atorvastatin can affect the intercellular interaction between endothelial cells and mesangial cells exposed to high glucose and LPC.

Objective we got a lot of recombinant human α-defensin-3 by genetic engineering method, and studied whether to increase ciprofloxacin antibiotic activity to resistant strains of Pseudomonas aeruginosa. Method RT-PCR retroviral human blood leukocytes of the total RNA, and PCR amplification of the target gene, cloned into pET32a (+) plasmid. Transformed into the recombinant E. coli and the affinity chromatography purified protein. By calculating HNP-3 and ciprofloxacin MIC value and FIC detection of recombinant protein biological activity. Results Construction of a pET32a (+) / HNP-3 original expression vector, and the expression of protein was determined the purpose of protein. The FIC value of combination of HNP-3 and ciprofloxacin is less than 1.0. Conclusion human α-defensin-3 enhanced ciprofloxacin antibiotic activity to resistant strains of Pseudomonas aeruginosa.

Objective To explore the regulative effect and mechanism of basic fibroblast growth factor (bFGF) to β-catenin in cardiac fibroblasts. Methods 3days SD rat cardiac fibroblasts (CFs) were isolated and cultured in normal condition and then in hypoxia/reoxygenation (Hy/R) condition. We use the third generation cardiac fibroblasts and divided into 6 groups, including the control group, the Hy/R control group, the 5, 10, 20 and 40μg/L bFGF group. The MTT method was used to investigate the proliferation of cardiac fibroblasts, The expression of β-catenin and apoptosis cell were detected and measured by laser confocal scanning microscope method and TUNEL method. Results MTT A 490 nm value in Hy/R groups was significant decreased compare with control group (P<0.05), with β-catenin protein decreased (P<0.05) and apoptosis increased (P<0.05). Compare with Hy/R groups the MTT A 490 nm value of the bFGF groups were significantly increased (P<0.05), the expression of β-catenin in bFGF group was increased significantly (P < 0.05), and the cell apoptosis was decreased in bFGF group (P<0.05). Conclusion The results indicate that bFGF can up regulates the expression of β-catenin in the cardiac fibroblast, facilitates cell proliferation and depresses cell apoptosis after Hy/R.

Objective: To evaluate the related factors for adverse maternal and fetal outcome among women with HELLP syndrome (hemolysis,H, elevated liver enzyme, EL, low platelet count, LP).Methods: We retrospectively analyze maternal and fetal medical records of pregnancy complicated HELLP syndrome from January 1, 2005 to December 31, 2009. Related factors included maternal age, parity, gestational age at diagnosis, adverse maternal and fetal outcomes. Statistical analysis was performed with the χ2 test analysis.Results: A total of 32 pregnant women with HELLP syndrome were reviewed. The incidence of HELLP or ELLP was 2.16% among the women with severe pre-eclampsia (32/1484). Gestational age at diagnosis was about 32 weeks and 4 days, with a peak frequency between the 27th and 37th gestational weeks. In about 84.4% of the cases, the HELLP syndrome develops before delivery. The rests develops within 48 h after delivery. The incidence of maternal complication, such as eclampsia, abruption placenta, postpartum bleeding, acute renal deficiency was higher among women with HELLP syndrome as compared with ELLP. The incidence of HELLP was significantly associated with a platelet nadir below 50x109/L (p<0.05). High incidence of fetal and neonatal death was present before 30 weeks of gestational age and with intrauterine growth restriction. High incidence of HELLP syndrome was also present in women without regular care during pregnancy. Conclusions The maternal adverse outcome of HELLP was higher than ELLP. There was a significant association between a platelet nadir below 50x109/L and the severity of HELLP. The perinatal mortality and morbidity was high before 30 weeks. Regular care during pregnancy may play an important role in prevention maternal and fetal adverse outcomes.

Parkinson's disease (PD) is a common neurodegenerative disease in the central neural system. Genetic and environmental factors induce microglia secrete more interleukin-1, which will cause dopaminergic neurons death through several mechanisms, such as promoting the synthesis of other pro-inflammatory cytokines, producing free radicals, increasing the intracellular calcium concentration, and inducing elevation of α-synuclein , and so on. It is a potential therapeutic target for Parkinson's disease.

Neural stem cells are charactered with proliferation,migration and differentiation. The mobilization of endogenous neural stem cells has been a new therapeutic strategy in the central lesion. Recent date suggest that the endogenous neural stem cells can be triggered by administration of several strategies, such as trophic factors and cytokines, environmental enrichment and exercise, hyperbaric oxygen, traditional medicine, and so on. In addition, other endogenous neural cell types usable for central repair are also observered.

Recent studies have shown that mesenchymal stem cells (MSCs) have immunomodulatory properties and tissue repairing potential. In the process of acute pancreatitis, MSCs suppress T cells and B cells and many kinds of inflammatory factors, and home to injuried pancreas to promote pancreatic tissue repairing. MSCs play an important role in pathophysiolgical progresses of acute pancreatitis. The review is on the effect of MSCs on acute pancreatitis.

Objective This paper aims to popularize preventive and treatment knowledge of diabetes for diabetics, enhance their self-management ability and slow the progress of diabetes and reduce its complications. Methods This paper applies the research method of giving intensive management to the health education for 90 diabetics, and comparing the change of their blood sugar, blood fat, glycosylated hemoglobin and life style before and after the intensive management. Results Through three-month's intensive management, there has been a distinct decrease in levels of patients' fasting blood sugar, two-hour postprandial blood sugar and glycosylated hemoglobin respectively (P<0.05). The irrational structure and intake of the three major nutrients have been significantly improved. There has also been a great increase in the amount of effective exercise. Conclusion The intensive management of the health education on diabetes is significantly important to prevent and cure diabetes.
