姜黄素提高人前列腺癌细胞系PC-3放疗敏感性

Curcumin enhances the radio-sensitivity of prostate cancer cell line PC-3

摘要: 目的 探讨姜黄素提高人前列腺癌细胞系PC-3放疗敏感性的分子机制。方法 将细胞分为对照组、放射组、姜黄素组和姜黄素联合放射组,通过MTT,RT-qPCR和流式细胞仪等方法,观察PC-3细胞增殖、凋亡和细胞周期的变化。结果 放射治疗和姜黄素组,其细胞存活率、细胞中PCNA mRNA和Cdc2 mRNA水平均低于对照组(P<0.05),而其S期和G2/M期细胞比例、caspase-3活性、caspase-9活性以及细胞凋亡率均高于对照组(P< 0.05);姜黄素联合放射治疗组,其细胞存活率、细胞中PCNA mRNA和Cdc2 mRNA水平均低于放射组(P<0.05),S期和G2/M期细胞比例、caspase-3活性、caspase-9活性以及细胞凋亡率均高于放射组(P<0.05)。结论 姜黄素具有一定放射增敏效应,可显著提高放射治疗对人前列腺癌细胞PC-3增殖的抑制效应。

Abstract: Objective To explore the molecular mechanism of curcumin increasing the radiosensitivity of prostate cancer PC-3 cells. Methods Prostate cancer PC-3 cells were cultured and treated with curcumin, irradiation or combined therapeutic methods to observe the proliferation, apoptosis and change of cell cycle of PC-3 cells shown by MTT,RT-qPCR and FCM. Results Compared with control group, the cells from the radiation or curcumin group, the survival rate, the level of PCNA mRNA and Cdc2 mRNA were decreased (P<0.05), and the ratio of S phase and G2/M phase, caspase-3 and caspase-9 activity and the apoptosis rate were increased (P<0.05); Compared with the cells in the radiation group, after treated with curcumin combined with radiotherapy, the survival rate, the level of PCNA mRNA and Cdc2 mRNA were decreased (P<0.05), and the ratio of S phase and G2/M phase, caspase-3 and caspase-9 activity and the apoptosis rate were all increased (P<0.05). Conclusions Curcumin has a certain radiosensitizing effect in vitro, which potentially strengthen the inhibitory effect of radiotherapy on the growth of prostate cancer PC-3 cells.
