甘露醇促进人肺鳞癌细胞系NCI-H292凋亡

摘要: 目的 探讨20%甘露醇注射液对人肺鳞癌细胞系NCI-H292增殖和凋亡的作用及其可能机制。方法 使用20%及以下不同浓度甘露醇处理NCI-H292细胞,CCK-8法检测NCI-H292细胞增殖和活性;流式细胞测量术检测细胞凋亡;Western blot检测线粒体凋亡途径相关蛋白Bcl-2和Bax的表达。结果 与对照组比较,甘露醇呈剂量和时间依赖性抑制细胞增殖(P<0.05); 20%甘露醇可诱导细胞凋亡(P<0.05);经20%甘露醇处理后的细胞中Bcl-2的表达水平均显著降低,Bax的表达水平均明显升高(P<0.05)。结论 临床上常用的20%甘露醇注射液可以诱导肺鳞癌细胞系NCI-H292凋亡,其机制与线粒体途径相关蛋白Bcl-2、Bax有关。

Abstract: Objective To investigate the effects of 20% mannitol injection on the proliferation and apoptosis of human lung squamous carcinoma cell line NCI-H292 and its possible mechanisms. Methods NCI-H292 cells were incubated with 20% and lower concentrations of mannitol. Cell proliferation and viability were detected by CCK-8 assay. Cell apoptosis was detected by flow cytometry and the expression of mitochondrial apoptosis pathway-related proteins Bcl-2 and Bax was detected by Western blot. Results Compared with the control group, mannitol inhibited cell proliferation in a dose- and time-dependent manner (P< 0.05), 20% mannitol could induce cell apoptosis (P< 0.05). The expression level of Bcl-2 was significantly reduced, while that expression level of Bax was significantly increased in cells incubated with 20% mannitol (P< 0.05). Conclusions Clinically used 20% mannitol injection can induce apoptosis of lung squamous carcinoma cell line NCI-H292. Its mechanism is related to mitochondrial pathway-related proteins Bcl-2 and Bax.
