胆汁淤积性肝损伤炎性反应机制的研究进展

Research progress on the inflammatory mechanisms of cholestatic liver injury

摘要: 胆汁淤积性肝损伤(CLI)是临床常见肝脏疾病,其生化本质是各种原因导致胆汁酸异常蓄积,使系统胆汁酸暴露增加,并在肝脏内造成肝细胞损伤。若病情持续进展,可以发展为肝纤维化、肝硬化、肝衰竭和肝癌等疾病。本文聚焦于胆汁淤积引发肝细胞损伤的生物学环节,从炎性反应的角度探讨胆汁淤积导致肝损伤的分子机制,将对中性粒细胞、巨噬细胞(Kupffer细胞)及胆管细胞等在CLI相关炎性反应中作用以及介导CLI相关炎性反应的信号通路进行阐述,为寻找新的治疗靶点和治疗策略提供思路。

Abstract: Cholestatic liver injury (CLI) is a liver disease with high mordity. Accumulation of bile acids in the liver is thought to be a major driver of liver injury during cholestasis. The disease can develop into be liver fibrosis, cirrhosis, liver failure and liver cancer. This review focuses on the molecular mechanism of CLI in the perspective of inflammatory response. The roles of neutrophils, Kupffer cells, bile duct cells, and the signaling pathways involved in CLI are reviewed to provide clues for research of new therapeutic targets and treatment strategies.
