过表达TGF-β1可抑制人宫颈癌细胞系C-33A增殖和促进凋亡

Overexpression of TGF-β1 inhibits proliferation and promotes apoptosis of human cervical cancer cell line C-33A

摘要: 目的 研究TGF-β1基因转染对人宫颈癌细胞系(C-33A)增殖和凋亡的影响。方法 取增殖状态良好的宫颈癌细胞C-33A,用基因转染的方法建立TGF-β1过表达质粒,用RT-PCR和Western blot法分别检测 TGF-β1 mRNA和蛋白的表达及其Bcl-2和Bax的表达;用MTT实验、Transwell小室迁移实验和流式细胞术分别检测细胞的增殖率、迁移率及凋亡率。结果 TGF-β1 mRNA和蛋白在宫颈癌C-33A细胞中的表达低于其它细胞(P<0.05);TGF-β1抑制Bcl-2的表达(P<0.05)、促进Bax的表达(P<0.05);过表达TGF-β1细胞的增殖与迁移速度低于其他细胞(P<0.05),但凋亡速度高于其他细胞(P<0.05),且这种降低与升高均呈时间依赖性。结论 过表达的TGF-β1可明显抑制宫颈癌细胞的增殖和促进凋亡。

Abstract: Objective To study the effect of TGF-β1 gene transfection on proliferation and apoptosis of human cervical cancer cells in vitro. Methods Overexpression plasmid of TGF-β1 was established by gene transfection in C-33A cell line with well proliferation status. The expression of TGF-β1 mRNA and protein, Bcl-2 and Bax were detected by RT-PCR and Western blot. Cell proliferation rate,migration rate and apoptosis were detected by MTT assay, Transwell assay and flow cytometry, respectively. Results The expression of TGF-β1 mRNA and protein in cervical cancer cells was lower than that of other cells (P<0.05). TGF-β1 could inhibit the expression of Bcl-2 (P<0.05) and promote the expression of Bax (P<0.05). The rate of proliferation and migration of overexpressed TGF-β1 cells was lower than that of other cells (P<0.05), but the rate of apoptosis was higher than that other cells (P<0.05), and the decrease and increase show time dependence. Conclusions Overexpression TGF-β1 can significantly inhibit the proliferation and promote apoptosis of cervical cancer cells.
