双酚A增加CD1小鼠肝细胞凋亡

Bisphenol-A increases hepatocyte apoptosis in CD1 mice

摘要: 目的 观察在双酚A(BPA)的作用下,CD1小鼠肝细胞凋亡是否受到影响。方法 48只4周龄雄性CD1小鼠随机分成4组(12只/组),分别予以含不同浓度BPA(0、5、50和500 μg/(kg?d))的标准饲料喂养8周后,利用HE染色、Hoechst 33258染色观察肝脏组织形态学及细胞凋亡,Western blot法检测肝脏组织中cleaved caspase3、Bax和Bcl-2蛋白表达,用分光光度法测定caspase3酶活性。结果 与对照组相比,BPA各处理组肝脏组织中凋亡细胞明显增多(P<0.01),肝脏组织促凋亡基因cleaved caspase3与Bax蛋白表达增加(P<0.05);caspase3酶活性显著增强(P<0.01)。结论 在环境内分泌干扰物BPA的作用下,CD1小鼠肝细胞凋亡增加。

Abstract: Objective To investigate the effect of BPA on hepatocyte apoptosis in CD1 mice. Methods Forty-eight 4-week old male CD1 mice were randomly divided into 4 groups. Mice from different groups were exposed to different doses of BPA (0, 5, 50, 500μg/(kg?d)) through food contamination for 8 weeks. After the decapitation of mice and the isolation of liver, HE staining and Hoechst 33258 staining were used to observe the hepatocyte apoptosis; Western blot was performed to determine the protein expression of cleaved caspase3, Bax and Bcl-2 in liver; caspase-3 activity was detected using a caspase-Glo3/7 assay kit. Results Compared with the control group, mice treated with different concentrations of BPA showed enhanced apoptotic cells and caspase-3 activity in liver(P<0.01), with significantly increased protein expression of cleaved caspase 3 and Bax(P<0.05). Conclusions BPA induces hepatocyte apoptosis in CD1 mice.
