Influenza (simply as the flu), a kind of acute respiratory infection, is a commonly-seen contagious disease in human, wild birds, swine, horse and other fowl. Its onset is soon, infects quickly and spreading wide. It is a great danger to people's lives. There have been epidemic records of influenza in each century since the 6th century, among which the 1918's caused the most severe consequence. This article contains three aspects. They are naming and classification of flu, epidemic history of flu, Avian Influenza and Swine Influenza. The progress in diagnosis, treatment and prevention on flu are also reviewed. The main purpose of this research is to profoundly reconsider the relationship between human and nature, human and animal as well as human and environment. We need to work hard to reach an ideal harmonious state.

Objective: To observe the changes in neurotrophic factors and their receptors in the lumbar spinal cord of age-increasing rats (1, 3, 6, 12, 18 and 24 months), and to investigate their correlation with age increasing. Methods: The expression of neurotrophic factors and their high receptors in the lumbar spinal cord of rats were measured by immunohistochemistry and western blot. Nissl staining was used for detecting the number of survival neurons. Results: The expression of nerve growth factor (NGF) and its receptor TrkA, brain derived neurotrophic factor (BDNF) and its receptor TrkB, and gliocyte derived neurotrophic factor (GDNF) in lumbar spinal cord of rats showed changes in relation to the age increasing. Their expression was lower at the age of 1 month, reached the peak in 6-month-old rats, and decreased significantly at the age of 24 months, in which the decline of GDNF was more obvious. The number of survival neurons in lumbar spinal cord kept at a high level from 1 to 6-month-old rats, then decreased gradually, and reached the lowest level at the age of 24 months. Conclusion: The expression of neurotrophic factors and their receptors shows changes with age increasing, decreases in aged rats especially, which may be one of the mechanisms involved in spinal cord aging.

Objective To investigate whether conventional protein kinase C (cPKC) II- interacting collapsin response mediator protein-2 (CRMP-2) could provide neuroprotection against cerebral ischemic (I) injuries. Methods Male BALB/c mice were randomly divided into normoxic control (Nor), HPC, Nor+Sham, HPC+Sham, Nor+I and HPC+I groups (n=6 per group). Using our HPC and MCAO mouse models, we applied immunoprecipitation, two-dimensional electrophoresis and mass spectrometry to characterize cPKC II-interacting proteins and combined with SDS-PAGE and Western blot to quantitatively analyze CRMP-2 phosphorylation and degradation levels in the brain of mice after HPC and MCAO. Results Proteomics analysis revealed that the expression levels of 10 cPKC II- interacting proteins changed obviously in cerebral cortex of HPC mice when compared with Nor group. One of these proteins, CRMP-2 protein level increased in particulate fraction and decreased in cytosolic fraction of cerebral cortex of HPC mice. CRMP-2 phosphorylation level in ischemic core (Ic) of cerebral cortex decreased significantly (p<0.05, n=6) as compared with that of Nor+sham group, but CRMP-2 phosphorylation levels in HPC+I group increased significantly compared with that of Nor+I group (p<0.05, n=6). In ischemic cortex, CRMP-2 degradation (proteolysis) was observed as the appearance of 55-ku breakdown products (BDP). However, the CRMP-2 degradation level, BDPs products decreased significantly in penumbra (P) of ischemic cortex from HPC+I group when we compared with that of Nor+I group (p<0.05, n=6). Conclusion CRMP-2 was involved in attenuating the decrease of CRMP-2 phosphorylation in ischemic core and inhibiting its degradation in penumbra of cerebral cortex of mice thereby to lessen the ischemic injuries.

Objective To explore the mutual effect of proinflammatory factor tumor necrosis factor(TNF)-α and cigarette smoke extract(CSE) in the induction of mucin 5AC in BEAS-2B human airway epithelial cells.Methods The BEAS-2B cells were transfected with nuclear factor(NF)-κB decoy oligonucleotides(ODNs) or transfected with NF-κB scrambled ODNs as negative control,then treated with TNF-α,CSE,and TNF-α plus CSE,respectively. The expression of phosphorylated epidermal growth factor receptor(p-EGFR),mucin 5AC peotein production and gene expression were detected by Western blot,ELISA and RT-PCR,respectively.Results It was showed an obvious increasing of p-EGFR protein production in scrambled ODNs transfected cells exposed to stimuli,with elevation of MUC5AC protein production and mRNA expression,and the TNF-α and CSE co-incubated groups were much higher than single TNF-α or CSE stimulated group(P<0.05).The relative content of p-EGFR protein,MUC5AC mRNA,and MUC5AC protein in the TNF-α and CSE coincubated groups were 0.74±0.06,0.77±0.10,and 0.72±0.03,respectively,compared with 0.58±0.08,0.52±0.07,and 0.53±0.11 in CSE-stimulated group and 0.53±0.04,0.58±0.11,0.56±0.05 in TNF-α-induced group,all P<0.05. In NF-κB decoy ODNs transfected cells,TNF-α stimuli revealed an obvious reduction of MUC5AC protein production and gene expression,were 0.35±0.12,and 0.41±0.12,respectively,compared with scrambled ODNs transfected group,P<0.05,but had little effect in CSE group and co-incubated group,which still at a relative high level, were 0.48±0.04,0.49±0.11 and 0.51±0.08,0.48±0.07,respectively,compared with negative control group,P<0.05. Conclusion TNF-α and CSE can synergistically induce mucin 5AC expression in BEAS-2B cells, transcription factor NF-κB plays a critical role in TNF-α induced mucin production but not as so important in CSE stimulated cells.

Objective To investigate the effects of the human S100A6 on β-catenin in human osteosarcoma cell lines MG63 and U2OS . Methods Cell lines MG63 and U2OS were infected by recombinant adenoviruses carrying the human S100A6 and its siRNA gene, AdS100A6 and AdSiS100A6, respectively, to up-regulate and down-regulate the expression of S100A6. Then RT-PCR,Western blot and immunocytochemistry were used to detect mRNA and protein (level and/or distribution ) of β-catenin. Results In both cell lines MG63 and U2O S, with up-re gulation of S100A6, the expression of β-catenin mRNA and protein increased(P<0.05)and β-catenin protein increase was more obvious in nuclear than in cytoplasma; while down-regulating S100A6, both the mRNA and protein level of β-catenin decreased(P<0.05); β-catenin protein decrease was more obvious in nuclear than in cytoplasma, too. Conclusion. Increaseing Wnt/β-catenin signaling activity may be a mechanism that S100A6 involves in tumor development.

Objective To study expression enhancement and purpose of Y14 and Upf1 in human breast cancer cell lines and human breast cancer tissue. Methods Immuocytochemistry and laser scanning confocal microscope(LSCM) were applied. Y14 and Upf1 were determined in human breast cancer cell lines(MCF-7、ZR-75-30、T47D、MDA-MB-435s、MDA-MB-453、MDA-MB-231)and breast epithelial cell line(HBL-100).Results ①Y14 and Upf1 levels of breast cancer cells are obviously higher than the breast epithelial cell line(P<0.05).②Y14 and Upf1 levels of MDA-MB-231 are obviously higher than MCF-7.③The expression enhancement of Y14 and Upf1 levels are obviously higher in human breast cancer tissue. Conclusion The expression levels of Y14 and Upf1 in the breast cancer cells and tissue enhance obviously.

Objective To construct a human phage single chain-antibody library, and sieve out the antibody ScFv against lung cancer from the library. Methods The total RNA was isolated from lymph node tissue of the lung cancer patients, and was used to amplify VH and VL gene by RT-PCR. VH and VL were joined with a DNA linker by SOE-PCR to form the single chain variable fragment (ScFv) gene. ScFv gene was coloned into the phage vector pCANTAB5E. Panning against lung cancer cell line A549 were performed for four rounds. The positive clones were chosen for soluble expression. Results A recombination phage single chain-antibody library was constructed. After 4 rounds panning, the number of eluted phages increased 115 times. Positive reactions to A549 were detected in 7 of 10 random clones, The positive rate was 70%. The human ScFvs against lung cancer were produced and confirmed by SDS-PAGE and ELISA analysis. Conclusions ScFvs against lung cancer were acquired by the construction of phage single chain-antibody library. The soluble ScFvs has avidity specifically to human lung cancer cells.

Objective To study the relationship between HLA-A alleles, supertype, HPV infection and cervical cancer in Tu Nationality of Hubei province.Methods in the same geographic region and of similar age .The comparisons included the comparison between HPV positive cases and HPV positive women in control group, and the comparison between HPV positive cases and HPV negative women in control group. Number of cases was 100( HPV positive was 86) , and control was 187 (HPV positive was 95 and HPV negative was 92). The most polymorphism of 2 and 3 exons of the HLA-A alleles were analyzed by the high-resolution typing method-sequence-based typing(SBT). Results Comparison between HPV positive cases and HPV positive control women. Supertype HLA-A3 (pcorrected=0.005, OR=2.36, 95%CI=1.45-3.85) was risk factors. Comparison between HPV positive cases and HPV negative control women, HLA-A*0206 alleles(pcorrected=0.025 ,OR=0.20,95%CI=0.07-0.58)Supertype HLA-A2(pcorrected=0.005, OR=0.57,95%CI=0.37-0.88)was protective factor. SupertypeHLA-A3 (pcorrected=0.005, OR=2.36, 95%CI=1.45-3.85) was also related to the susceptibility of cervical carcinoma.Concusion SupertypeHLA-A3 was risk factor of cervical cancer.

Objective To investigate the distribution of the emm types and superantigens of group A streptococcal (GAS) isolated from Chinese children. Method 222 GAS isolates obtained from five Children's Hospital of China during 2005-2006 were studied, emm types were performed by PCR and sequencing. And the eight superantigen (SAg) genes (speA, speC, speH, speI, speG, speJ, ssa, and SMEZ) were detected by PCR. Results Nine emm types were determined,of which emm12 (55.86%) and emm1 (39.64%) were the most prevalent types. The GAS isolates carried six or more than six SAg genes were 78.39% of all the isolates in this study. The SAg gene profiles were closely associated with the emm type. Conclusion This study showed that the distribution of the emm types of S. pyogenes isolated from Chinese children were intensively. The SAg genes appeared to be associated with the emm type, thereby aiding in vaccine development in China.

Objective To discuss whether CX3CR1 V249I and T280M are associated with CAD. Methods Database in English and Chinese, including PUBMED, EMbase, were searched to get the case-control studies on the association between V249I, T280M and CAD. Results Six studies were enrolled. The pooled OR of V249I compared to wild type allele was OR=0.85(95%CI=0.67~1.08,P>0.05). The pooled OR of T280M compared to wild type allele was OR=0.82(95%CI=0.70~0.96,P<0.05). Conclusion The T280M is a protective factor of CAD, while the V249I is not associated with CAD.

Objective To analyze the characteristic of serum proteins in non-small cell lung cancer ( NSCLC) patients, establish serum markers pattern for the diagnosis of NSCLC. Methods Surface enhanced laser desorption ionization time of flight mass spectormetry( SELDI-TOF-MS ) technology was used to analyze serum samples including 100 cases of NSCLC patients and100 cases of healthy controls. Biomarker Pattern Software (BPS) was used to detect the protein peaks significantly different between NSCLC patients and healthy controls. And then established a diagnostic pattern.This pattern was further valuated by a blind test. Results Sixteen significantly different protein peaks were found in serum samples between NSCLC patients and healthy controls. Eight up-regulated protein peaks and eight down-regulated protein peaks (P<0.001)were identified in serum samples of patients of NSCLC comparing with healthy controls.Using BPS, a diagnostic pattern was established with 100% sensitivity and 100% specificity . A blind test generated a sensitivity of 96.15% and specificity of 95.83% respectively. Three up-regulated protein peaks(P<0.05)were identified in serum samples of patients of NSCLC with smoking history comparing with no smoking history. Two up-regulated protein peaks(P<0.01)were identified in serum samples of patients of squamous carcinoma comparing with adenocarcinoma .No significantly different protein peak was found in serum samples of NSCLC patients with different clinical stages. Conclusion SELDI-TOF-MS technology can identify the significantly different protein peaks and establish a diagnostic pattern with high sensitivity and specificity.It will provide a highly accurate approach for the diagnosis of NSCLC.

Objective To investigate the protective effects of aspirin on hypoxic brain neural cells of rat,and the effects of extracellular calcium on the neuroprotective effect of aspirin.Methods The hypoxic cell model was established by adding Na2S2O4 to the calcium or calcium free medium. Cultured primary cortical neurons of rat were pretreated with ASA in vitro.The change of intracellular free calcium and neuroglobin (NGB) were observed and analyzed by laser scanning confocal microscope. Results The expression level of [Ca2+]i and NGB increased significantly in the chemical hypoxia group(P<0.05).ASA can attenuate the increase of [Ca2+]i and NGB in the hypoxic group and calcium-free hypoxia group(P<0.05).Conclusion Aspirin can inhibit calcium overload and inhibit the hypoxia-induced expression of NGB, and protect rat brain cells.

Objective To investigate the effects of Ligustrazine on TNF-α-induced TGF-β1 and CTGF expressions in human peritoneal mesothelial cells (HPMCs). Methods HPMCs were isolated from human omenta by trypsin digestion method and subcultured. Then, the HPMCs were divided into control group, TNF-α-induced (1 μg/L) group and TNF-α-induced plus low-, medium- and high-dose Ligustrazine (10, 20 and 40 mg/L Ligustrazine respectively) groups. The viability of HPMCs was measured by MTT assay. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expressions of TGF-β1 and CTGF mRNAs in HPMCs. Proteins of TGF-β1 and CTGF in culture supernatants were measured by enzyme linked immunosorbent assay (ELISA). Cell protein concentration was measured by trace bicinchoninic acid (BCA) method to correct the ELISA assay results. Results Ligustrazine could significantly decrease TNF-α-induced TGF-β1 and CTGF expressions in a dose-dependent manner both in protein and gene levels (P<0.05). In addition, medium- and high-dose Ligustrazine injection could significantly ameliorate the viability of HPMCs inhibited by TNF-α (P<0.05). Conclusion Ligustrazine could inhibit expressions of TGF-β1 and CTGF in HPMCs cultured under inflammatory conditions.

Objective Analysing the association of mismatch repair protein MLH3 gene C2531T mutation in oligospermia and azoospermia cases. Methods Using MLH3 C2531T as an example to establish a method of tetra-primer amplification refractory mutation system-PCR (4P-ARMS-PCR);We chose 81 cases of oligospermia, azoospermia patients and 80 normal controls , detecting MLH3 C2531T both using 4P-ARMS-PCR and PCR-RFLP; randomly to select 20% samples to sequence. Results The date suggested higher prevalence of CT+TT genotypes in patients as compared to the controls and association of T allele with disease(P<0.01) ; PCR-RFLP results, 4P-ARMS-PCR results and sequence results are fully consistent. Conclusion 4P-ARMS-PCR can be a rapid, simple and accurate detection of gene mutation of single nucleotide polymorphism (SNP); MLH3 C2531T polymorphism is associated with oligospermia and azoospermia.

Objective Explore the polymorphism distribution of the hormone-sensitive lipase (HSL) gene i6 in ZunYi region. Methods Examine the polymorphism of the hsl i6 in the ZunYi population with ploymerase chain reaction- single strand conformation polymorphism (PCR-SSCP), and. analyzing the distribution of it. Results 6 different alleles and 11 different genotypes were found in the hsl i6. The PIC is more than 0.7, in which allele 6 and allele 7 have the most widely distributed Conclusion The polymorphism of hsl i6 was distributed in ZunYi.. And it provides the relevant for hsl as a candidate gene of glucose and lipid metabolism.

Objective To investigate the therapeutic effects of atorvastatin on pulmonary fibrosis of rats induced by Bleomycin(BLM). Methods 42 female Wistar rats were randomly divided into 7 groups: the control group(group C), the model group which was furtherly divided into group 2-week(M2), 4-week(M4), 6-week(M6), and atorvastatin-treatment group which was furtherly divided into group 2-week(A2), 4-week(A4), 6-week(A6). Each had 6 rats. Group M and A were induced to pulmonary fibrosis by the method of BLM endotracheal injection, while the group C was injectted with the saline. On 2nd day, group A were given orally atorvastatin by 10mg/ kg. d. Rats were seperately killed on 2nd, 4th and 6th week. After intratracheal injection of BLM. Alveolitis and pulmonary fibrosis were evaluated by pathology, hydroxypmline concentration and PaO2. Results The lung coefficient of group M2, M4, A2 and A4 was higher significantly than group C. The degree of pulmonary fibrosis in group A4 and A6 was improved compared with group M4, M6. Alveolitis and pulmonary fibrosis in group A were improved compared with those in group M.Hydroxypmline concentration in group A and M were significantly higher than that in group C. while A4 was lower than M4 (P<0.01). There was a increase in PaO2 after exposure to BLM, especially the group M2 (P<0.01). Improvement in PaO2 was observed in groups A, compared with M. Conclusion Atorvastatin had effective treatment on pulmonary fibrosis of rats. The effect was significant on 6th week after treatment, which suggest that the effect maybe correlated positively to the treatment duration.

Objective To explore the effect of Angiopoietin-2(ANG-2) and Endostatin (ENS) in genesis and development of endometriosis.Methods 25 endometriosis cases were enrolled into present study.24 patients without endometriosis were selected control. ANG-2 and ENS in endometrium were detected by Immunohistochemistry. The concentrations of ANG-2 and ENS in peritoneal fluid were determinated with enzyme linked immunoabsorbant assay.Results In the EMs group ,the ANG-2 positive rate in eutopic endometrium was significantly higher than that of ectopic endometrium . The ANG-2 positive rate in ectopic endometrium was significantly higher than that of control group;In EMs group, The ENS positive rate in ectopic endometrium was significantly higher than that of eutopic endometrium .The ENS positive rate in eutopic endometrium was significantly higher than that of control group. There was no significant difference of ANG-2 or ENS between stageⅠ~Ⅱand stage Ⅲ~Ⅳ. In peritoneal fluid, ANG-2 and ENS concentration and ratio of ANG-2/ENS in endometriosis were significantly higher than that in controls. Conclusion The effect of angiopoietin-2 and endostatin in angiogenesis of endometriosis are important and adjust to pathogenesis and development of the endometriosis .

The Results group HF than the group C, myocardial cell injury obviously, and apoptotic bodies can be seen, serum CPK, CK-MB and increased LDH,group LS than the group HF, myocardial apoptosis index lower (P <0.01), myocardial cells Bc1-2 expression, Bax expression reduction.Conclusion Losartan can effectively inhibit the process of heart failure occurred in cardiomyocyte apoptosis, reversal of myocardial damage and improve the process of the role of heart failure.

Objective To determine the role of arginase inhibitor (nor-NOHA) on vascular remodeling in SHRs。Methods WKY rats were used as the control and SHR were randomly divided into untreated control(SHR) and treated group(SHR-T). SHR in the treated groups were injected intraperitoneally every day with nor-NOHA(40 mg/kg )for 10 weeks while the control groups were given to saline. Indirect tail-cuff systolic blood pressures and body weight were measured at given times. After the experiment ,The thoracic aorta、left ventricular(LV) and left kidney were taken . The contents of activated MMP-2 in these organs were measured by semi-quantitative gelatine-zymography,meanwhile Media thickness(MT), luminal diameter(LD), ratio of media to lumen (M/L) and collagen type I and Ⅲ, elastic fibers of thoracic aorta were measured by special Tricrome Masson, Picric-Sirius red and Orcein staining methods. Results (1) Blood pressure of treated group was significantly lower than untreated control groups;(2) Much more MMP-2 in LV exsited in SHR group than in SHR-T group .(3)MT and M/L of treated group were significantly smaller than untreated control groups in thoracic aorta; Much more collagen I and III existing in the vascular of untreated control group compare with treated group.;Conclusions nor-NOHA could relieve vascular remodeling probably by inhibiting the activity of MMP-2 and the synthesis of collagen I and III.

Angiocentric Glioma has been newly identified as an epilepsy associated tumor recently. Up to now, such cases that have been reported are still scarce. The tumor causes drug-resistant epilepsy. On histological findings, monomorphous bipolar tumor cells are arranged around vessels. Imaging studies show that the tumor is diffused or rimlike hyperintensity on T1-weighted SE sequences and a stalklike extension to the ventricle on T2-weighted sequences. The World Health Organization listed this new entity in its fourth edition of the WHO classification of tumors of the central nervous system published in 2007.
