Changes of NCX and SERCA2a gene expression in myocardial tissues in rats with pressure overload-induced left ventricular

Abstract: Objective To observe the change of NCX,SERCA2a gene expression in myocardial tissues and abnormity of heart function in rats with pressure overload-induced left ventricular hypertrophy. Methods cardiac hypertrophy was induced by clipping the abdominal aorta in rats. The parameters of hemodynamics including LVSP,LVEDP,±dp/dtmax,and HR were measured.The cardiac hypertrophy was evaluated by Left ventricular index(LVWI,left ventricular weight/body weight). NCX,SERCA2a mRNA and protein expressions in left ventricular tissues were determined by half-quantitative RT-PCR and Western blot normalized to abundance of GAPDH mRNA and protein,respectively. Result LVSP and LVEDP were obviously enhanced(P<0.01,P<0.001) in rats with cardiac hypertrophy in vivo. LVWI was increased remarkably(P<0.001)in model group. RT-PCR and Western blot results demonstrated that NCXmRNA expression was upregulated(P<0.05),and SERCA2a mRNA and protein expression were downregulated(P<0.05) significantly in model group compared with sham group. Conclusion Abnormity of heart function,especially diastolic function was observed. in rats with pressure overload-induced cardiac hypertrophy,and that may be relative to down-regulation of SERCA2a expressions in left ventricular tissues.
