人II型CD74基因胞外端片段在昆虫细胞中的表达

Expression of hCD74 truncated fragment in baculovirus vector system

摘要: 目的 建立昆虫杆状病毒表达系统表达人II型 CD74胞外端片段的技术平台。方法 运用分子克隆技术,分别将CD 74胞外端全长(73-232 aa)和与MIF结合的核心区(109-192 aa)片段置于表达载体pFastBacDual的PPH启动子下,重组质粒转入含有Bacmid的大肠杆菌DH10Bac后与病毒基因组重组,提取重组病毒杆粒基因组转染Sf-21细胞系。用RT-PCR检测目的基因mRNA,SDS-PAGE和Western-blot分析重组蛋白的表达。结果 CD74胞外端全长(73-232 aa)可实现在昆虫细胞中的表达,目的蛋白可被His标签抗体和CD74抗体识别;在细胞感染72h,病毒接种滴度MOI为8时,目的蛋白可实现最大表达量。CD74与MIF结合的核心区109-192 aa片段在mRNA可实现转录的情况下未实现目的蛋白表达。结论 成功建立人II型 CD74胞外端全长在昆虫细胞Sf-21中的表达平台,并获得了目的蛋白表达的优化条件。

Abstract: Objective To establish an expression system of human II CD74 extracellular fragment by insect baculovirus vector expression system in insect cells. Methods hCD74 full extracellular fragment (73-232 aa) and the core area combined with MIF (109-192 aa) were amplified by PCR, and inserted into pFastBacDual vector placed promoter of PPH. Recombinant plasmid was transformed into E. coli DH10Bac and obtained recombinant virus bacmid by site specific transposition. Recombinant bacmid was transfected Sf-21 cell line. The target mRNA transcription were tested by RT-PCR and recombinant protein expression were analyzed by SDS-PAGE and Western-blot. Results The full extracellular fragment (73-232 aa) of hCD74 could be expressed in insect cells and the target protein can be detected using antibodies of His tag and CD74. In addition, it can achieve the maximum expression in cells after 72h infection and inoculation using the virus titer MOI 4. The core area of 109-192 aa fragment detected the mRNA transcription, but not achieve the protein expression. Conclusion hCD74 extracellular fragment was successfully expressed in Sf-21 cell, and optimized the best protein expression conditions.
