氯喹对人宫颈癌细胞系HeLa增殖、凋亡及自噬的影响

Effects of chloroquine on proliferation, apoptosis and autophagy of human cervical cancer cell line HeLa

摘要: 目的 探讨氯喹(CQ)对人宫颈癌细胞系HeLa增殖、凋亡及自噬的影响,并初步探讨其作用机制。方法 将HeLa细胞分为对照组(不加药物)和实验组(氯喹-1组至氯喹-5组,分别用25、50、75、100、150 μmol/L CQ处理)。细胞均培养24和48 h;CCK-8法和集落形成实验检测细胞增殖;活性氧检测试剂盒检测细胞内ROS产生;流式细胞测量术检测细胞凋亡率。蛋白免疫印迹检测自噬蛋白p62、LC3和Beclin1,凋亡蛋白Bax、Bcl-2和PARP以及PI3K/AKT/MDM2通路蛋白的表达水平。结果 CQ呈时间和浓度依赖性抑制细胞增殖(P<0.05);CQ显著抑制细胞自噬活性,诱导胞内ROS的产生,诱导细胞凋亡(P<0.05)。与对照组相比,CQ可明显抑制PI3K/AKT/MDM2信号通路的激活(P<0.05)。结论 氯喹可能通过PI3K/AKT/MDM2通路抑制人宫颈癌HeLa细胞的增殖,诱导细胞凋亡。

Abstract: Objective To investigate the effect of chloroquine (CQ) on proliferation, apoptosis and autophagy of human cervical cancer cell line HeLa and to preliminarily explore its mechanism of action. Methods HeLa cells were divided into control group and experimental groups: chloroquine-1 to chloroquine-5 groups (25,50,75,100, 150 μmol/L CQ respectively). The cells all were cultured for 24 and 48 h; CCK-8 and colony formation assays were used to detect cell proliferation; reactive oxygen species detection kit was used to detect intracellular ROS production; flow cytometry was used to detect apoptosis rate. The expression level of autophagy proteins p62, LC3 and Beclin1, apoptosis proteins Bax, Bcl-2 and PARP and PI3K/AKT/MDM2 pathway proteins was detected by protein immunoblotting. Results CQ inhibited cell proliferation in a time- and concentration-dependent manner (P<0.01); CQ significantly inhibited cell autophagy, induced intracellular ROS production and induced apoptosis (P<0.01). Compared with control group, CQ significantly inhibited the activation of PI3K/AKT/MDM2 signaling pathway (P<0.05). Conclusions Chloroquine may inhibit the proliferation of HeLa cells and induce apoptosis through the PI3K/AKT/MDM2 pathway.
