由ATRA诱导表达的Sp100对白血病NB4细胞增殖的影响

Expression of Sp100 induced by ATRAand its effecton proliferation in NB4 cells

摘要: 目的 探讨Sp100在全反式维甲酸(ATRA)作用NB4细胞过程中的表达及其对NB4细胞增殖和周期的影响。方法 实时定量PCR检测Sp100 mRNA的表达;Western blot 检测Sp100蛋白的表达;免疫荧光检测Sp100的定位;CCK-8检测NB4细胞的增殖;流式细胞术检测NB4细胞的周期。结果 ATRA能明显促进NB4细胞中Sp100mRNA水平和蛋白水平,并将Sp100由弥散的小点状转变为较大的斑块状;感染Sp100-shRNA的NB4细胞增殖活力明显高于未感染组和病毒空载组(p<0.05),并使G2/M期的细胞增多。结论 ATRA促进NB4细胞中Sp100的表达,Sp100可能参与NB4细胞增殖活力的调节。

Abstract: Objective To investigate the expression of Sp100 in ATRA-treated NB4 cellsandits effect on proliferation in NB4 cells.Methods Q-PCR was employed to measure the expression of Sp100 mRNA;Western blot was used to detectthe expression of Sp100 protein; Immunofluorescence was adopted to determine the location of Sp100;Cell viability was analyzed by CCK8;Flow cytometry was used for cell cycle analysis. Results ATRA could induce obviously the expression of mRNA and protein of Sp100. ATRA changed the location of Sp100 from a micro-punctate pattern into a punctate nuclear patternin NB4 cells. Sp100-shRNA promoted the proliferation of NB4 cells and increased the cells in G2/M phase.Conclusions The expression of Sp100 was significantly increased in ATRA-treated NB4 cells, and Sp100 may be involved in the regulation of proliferation activity of NB4 cells.
