过表达Rab27B体外促进人胃癌细胞系MKN-45凋亡并抑制其增殖

Over-expressed Rab27B induces apoptosis and inhibits proliferation of MKN45 cells in vitro

摘要: 目的 探讨Rab27B在胃癌细胞株MKN-45增殖和凋亡中的作用。方法 构建Lv-Rab27B-EGFP表达载体并转染细胞作为实验组,以Lv-EGFP空载体转染细胞作为对照组,正常MKN-45作为空白组,CCK8 测定细胞的增殖,流式细胞术检测细胞的周期和凋亡, Western blot 检测凋亡相关蛋白 Bax、 Bcl-2 和caspase3的表达。结果 与阴性对照组和空白组细胞比较,实验组细胞的增殖受到抑制(P<0.05),细胞周期显示有丝分裂受阻于G1期,出现凋亡增多(P<0.05)。与对照组比较,实验组细胞促凋亡蛋白 Bax 表达量增加(P<0.05),抑凋亡蛋白Bcl-2 表达量显著下降(P<0.05),caspase3蛋白也明显升高(P<0.05)。结论 Rab27B过表达可以调节细胞周期而抑制人胃癌细胞株MKN-45增殖,要通过上调 Bax 和caspase3蛋白,以及下调 Bcl-2 蛋白表达来促进细胞凋亡。

Abstract: Objective To explore the effects of Rab27B on the proliferation and apoptosis in human MKN-45 cells. Methods Constructed the Lv-Rab27B-EGFP carrier and transferred it to MKN-45 cells as Rab27B group. Set up blank and negative control group. Cell growth was measured by CCK8 assay, the cell cycle and apoptosis were detected by flow cytometry. Relative expression levels of Bax, Bcl- 2 and caspase3 were detected by Western blot. Results Compare to blank and negative control cells, the proliferation of Lv-Rab27B-EGFP MKN-45 cells were decreased(P<0.05). Flow cytometry showed that over-expressed Rab27B resulted in an increase of G0/G1 phase and apoptotic cells. Expression of pro-apoptotic protein Bax was increased, and that of anti-apoptotic protein Bcl-2 was decreased differentially(P<0.05), the expression of caspase3 also showed significant difference(P<0.05). Conclusions Rab27B over-expression can inhibit cell proliferation by regulating cell cycles and induce apoptosis mainly through regulating Bcl-2 and caspase family proteins expression in MKN-45 cells.
