西南地区类风湿性关节炎患者Fas单核苷酸多态性与血清sFas水平相关

Assiociation between Fas polymorphisms and serum sFas level with rheumatoid arthritis patients in southwest China

摘要: 目的 探索中国西南地区人群Fas的rs2234767和rs1800682单核苷酸多态性(SNP)与类风湿性关节炎(RA)及其外周血血清可溶性Fas(sFas)水平的关系。方法 用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)检测183例RA患者和195名健康人群Fas的rs2234767和rs1800682多态性,采用酶联免疫吸附法(ELISA)检测外周血sFas水平,并对结果进行统计学分析。结果 RA患者和健康人群Fas的rs2234767和rs1800682多态性差异无统计学意义,RA患者外周血sFas蛋白水平[(2.54±0.41)ng/mL]显著高于健康对照组[(1.57±0.36)ng/mL](P<0.01),健康人群外周血sFas水平和Fas多态性无显著关联,RA组的Fas -670 G/G纯合子型的sFas水平[(2.37±0.29)ng/mL]较A/A型[(2.64±0.33)ng/mL]低(P<0.05)。结论 中国西南地区人群Fas的多态性与类风湿性关节炎的发生并无显著关系,但RA患者rs1800682基因型与外周血sFas水平有一定关联。

Abstract: Objective To clarify the association between Fas gene polymorphisms and the level of soluble Fas(sFas) in the serum of rheumatoid arthritis (RA) patients in southwest China. Methods Fas rs2234767 and rs1800682 single mucleotide polymorphisms(SNP) were genotyped in a case-control study with 183 RA patients and 195 controls, using polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP). sFas level in peripheral blood of RA patients and healthy people with different genotypes were detected by ELISA. Results There was no statistically significant difference between the patients with RA and the healthy subjects with respect to Fas rs2234767 and rs1800682 genotype distribution. However, patients carrying the rs1800682G/G genotype had a significantly decreased sFas level in serum compared with those carrying the AA genotype (P<0.05). Conclusions Fas polymorphisms do not represent a major genetic risk factor for RA susceptibility. However, Fas rs1800682 polymorphism may influence sFas level in serum and response in patients with rheumatoid arthritis of Chinese population in southwest.
