miR-195-5p表达上调抑制肺癌细胞系A549的迁移和侵袭

摘要: 目的 探讨miR-195-5p对肺癌细胞系A549细胞迁移和侵袭的影响及其机制。方法 实时荧光定量PCR检测肺癌组织及肺癌细胞系中miR-195-5p的表达;通过脂质体介导将miR-195-5p模拟物作用于A549细胞,用Transwell法检测细胞迁移和侵袭;Western blot检测细胞内上皮间质转化标志物及ZEB1的表达。结果 相对于癌旁组织及人正常肺上皮细胞,miR-195-5p在肺癌组织及肺癌细胞系中低表达(P <0.001);过表达miR-195-5p可显著抑制A549细胞的迁移和侵袭(P <0.001),伴随E-cadherin蛋白表达的上调,N-cadherin和vimentin蛋白表达的下调(P <0.01);过表达miR-195-5p可抑制A549细胞ZEB1的表达(P <0.001)。结论 miR-195-5p可能通过下调ZEB1的表达,抑制肺癌细胞迁移和侵袭。

Abstract: Objective To investigate the effect and mechanism of miR-195-5p on the migration and invasion of A549 cells in vitro. Methods The expression of miR-195-5p in lung cancer tissues, paracancerous tissues and lung cancer cell lines were analyzed by quantitative real-time PCR. Moreover, miR-195-5p mimics were transfected into A549 cells by liposome-mediated method. The cell migration and invasion were examined by Transwell assay. Western blot analysis was used to detect the effect of miR-195-5p on the protein level expression of epithelial-mesenchymal transition makers and ZEB1. Results Compared with paracancerous tissues and normal lung epithelial cells, the expressions of miR-195-5p were lower in lung cancer tissues and lung cancer cell line(P <0.001). The over-expression of miR-195-5p inhibited the migration and invasion of A549 cells(P <0.001), increased the protein expression of E-cadherin and reduced expression of N-cadherin、vimentin and ZEB1(P <0.01). Conclusions miR-195-5p inhibits the migration and invasion of lung cancer cells by reducing the expression of ZEB1.
