摘要: 目的 探讨肝细胞黏附分子(HEPACAM)基因转染肾癌细胞对其凋亡的影响。方法 将携带HEPACAM基因的重组质粒转染786-0细胞,RT-PCR鉴定HEPACAM基因在786-0中的表达; Hoechst染色结合荧光显微镜观察细胞形态改变;FCM检测细胞周期及细胞凋亡情况;Western印迹法检测BAX、BCL-2、CYCLIN D1 蛋白表达变化。结果 上调HEPACAM基因表达可诱导786-0细胞凋亡阻止细胞周期于G0/G1期,并可上调BAX及下调BCL-2和CYCLIN D1蛋白水平表达。结论 外源导入HEPACAM基因可诱导肾癌细胞凋亡并阻止细胞周期于G0/G1期。

Abstract: Objective To investigate the effect of transfection of hepatocyte cell adhesion molecule(HEPACAM) on apoptosis of renal cell carcinoma cells . Methods The vector containing HEPACAM gene was transfeceted into 786-0 cells, the expression of HEPACAM gene in 786-0 cells was confirmed by RT-PCR; The morphological changes of nucleus was observed by hoechst staining; The proliferation and apoptosis of 786-0 cells were analyzed by FCM; The expression of BAX、BCL-2、CYCLIN D1 were measured by western blotting. Results Up-regulated the expression of HEPACAM gene in 786-0 cells can arrest cell cycle at G0/G1 phase and induce cell apoptosis, and the expression of BAX was increased and the expression of BCL-2 and CYCLIN D1 was decreased. Conclusion These data indicated that ectogenic transfected HEPACAM gene may induce cell apoptosis and arrest cell cycle at G0/G1 phase in renal cell carcinoma cells.
