白细胞分化抗原151促进人结直肠癌细胞系HT29的增殖及侵袭

CD151 promotes the proliferation and invasion of colorectal cancer cell line HT29

摘要: 目的 研究白细胞分化抗原151(CD151)对人结直肠癌细胞系HT29增殖、侵袭能力的影响及其机制。方法 将细胞分为对照组细胞(HT29细胞)及CD151稳定敲低组细胞(CD151-HT29细胞)。RT-qPCR检测细胞中CD151的mRNA表达;蛋白印迹检测细胞中CD151、LRG-1、LGR-5以及β-catenin蛋白的表达;MTS实验、集落形成以及Transwell小室法实验检测细胞的增殖以及侵袭能力。结果 CD151-HT29细胞中CD151的表达较对照组细胞明显降低(P< 0.05); CD151-HT29细胞的增殖能力、集落形成能力以及侵袭能力较对照组细胞明显降低(P< 0.05);CD151-HT29细胞组的LRG-1、LGR-5以及β-catenin蛋白表达较对照组细胞明显降低。结论 CD151可促进结直肠癌细胞HT29的增殖及侵袭。

Abstract: Objective To study the effect of CD151 on proliferation and invasion of colorectal cancer cell line HT29 and the correlated mechanism. Methods The cells were divided into HT29 cell group and CD151-HT29 cell group. The expression of CD151 was detected by RT-qPCR; the protein expression of CD151, LRG-1, LGR-5 and β-catenin was detected by Western blot. The proliferation and invasiveness of HT29 and CD151-HT29 cells were observed by MTS proliferation, cloning and Transwell chamber assay. Results The expression of CD151 in CD151-HT29 cells was significantly lower than that in HT29 cells (P< 0.05); the proliferation and invasion ability of CD151-HT29 cells were significantly less than HT29 cells (P< 0.05). The protein expression of LRG-1, LGR-5 and β-catenin in CD151-HT29 cells was significantly lower than that in HT29 cells. Conclusions CD151 can promote the proliferation and invasion of colorectal cancer.
