摘要: 核酸适配体(aptamer)是一类能以较高的亲和力与各类生物靶标特异性地相结合的单链DNA或RNA,它们可以通过指数级富集的配体进化技术(SELEX)获得。与抗体相比,核酸适配体具有能够大量化学合成、低免疫原性、容易修饰、价格低廉等优点。核酸适配体技术和纳米及基因干扰技术的结合,为肿瘤的分子诊断和治疗提供了全新的思路和技术路径。本文就近些年来核酸适配体在肿瘤学方面的一些重要应用研究做一综述。

Abstract: Aptamers are single stranded DNA or RNA molecules that can specifically bind to their targets with high affinity. They are generated by a procedure called SELEX (systematic evolution of ligands by exponential enrichment). Comparing to antibodies, aptamers possess distinctive advantages: ability for chemical synthesis in large quantities, low-immunogenicity, easy chemical modification and low synthesis cost. Combining nanobiotechnology and RNAi with aptamers, this technology opens the way to more sophisticated applications in tumor molecular diagnosis and therapy.
