Adenovirus-mediated overexpression of BMP9 inhibits proliferation and migration in human osteosarcoma cell line 143B

Abstract: Objective The aim of this study is to investiage the effect of bone morphogenetic protein 9(BMP9) on the biological behavior of human osteosarcoma cell line 143B.Methods Using RT-PCR and Western blot annlysis to characterize the endogenous expression of BMP9 in the osteosarcoma cell line. Using a recombinant adenovirus expressing BMP9(adBMP9)to infect the osteosarcoma cell line with relatively low endogenous BMP9,then Transwell assay was used to determine cell invasion; Wound-healing assay was used to determine cell migration;MTT assay and crystal violet staining were used to determine the influence of adBMP9 on osteosarcoma cell proliferarion; Hoechst 33258 assay was used to determine cell apoptosis;Clonoy formation assay was used to determine cell clonoy. Results Osteosarcoma cell proliferation、migration、invasion and colony formation were significantly decreased while the cell apoptosis was induced by adBMP9. Conclusion adBMP9 is able not only to inhibit cell proliferation and migration but also induce cell apoptosis in human osteosarcoma cell line 143B.
