Expression of related cytokines of peripheral regulatory, CD4+effector T cells in patients with type 1 diabetes

Abstract: Objective Analyze and compare the amount of regulatory T cell and CD4+effector T cell, expression of IL-2, IL-10, IL17 and IFN-gamma in patients with type 1 diabetes (T1DM).Methods 45 T1DM, 37 T2DM patients (age ≤45 years, duration of the disease≤5 years, BMI≤30kg/m2) and 35 healthy subjects were involved. The amount of regulatory T cell and CD4+ effector T cell was analyzed by flow cytometry. Expression of cytokines such as IL-2, IL-10, IL17 and IFN-gamma were measured by real time-qPCR. Results There was negative correlation between Treg percentage and IFN-gamma expression in T1DM group (rs=-0.678, P=0.001). Positve correlation between Treg percentage and IL-10 expression was viewed in T1DM group (rs=0.773,p<0.001). Conclusion Out-of-balance of IL-10 and IFN-gamma in type 1 diabetes group suggested that the balance between preventive and destructive factors was broken.
