Involvement of caspase-3 activation and PARP-1 inhibition in DMSO induced apoptosis in A549 cells

Abstract: Objective To detect whether Poly(ADP-ribose) polymerase-1(PARP-1) is cleaved and therefore inhibited. Methods TUNEL assay and MTT assay of Caspase-3 activity were used to detect apoptosis; and PARP-1 cleavage was detected by western blotting. Results DMSO induced apoptosis in A549 cells and caspase-3 was activated in induced apoptosis, whereas PARP-1 was cleaved into two fragments. Conclusion During DMSO-induced apoptosis in A549 cells, we found that as caspase-3 was activated, PARP-1 was inhibited.
