Dexamethasone inhibits expression of aquaporin-4 of non-pigmented ciliary epithelium in rabbit

Abstract: Objective To study the effects of dexamethasone (Dex) on the expression of aquaporin-4(AQP4) in rabbit non-pigmented ciliary epithelium(NPCE). Methods Firstly to construct rabbit corticosteroid-induced glaucoma model and then contrast the differences of AQP4 expression in NPCE with normal controls by immunohistochemistry and immunofluorescence methods, as well as western blot method. Results The immunohistochemistry tests indicated that the AQP4 expression in NPCE of DEX group (OD=0.119±0.072) was suppressed than normal controls (0.242±0.056) (p<0.05). After some time the IOP decreased some of above changes gradually recovered (0.119±0.072), so did the western blot tests. Conclusions DEX increased IOP by changing the expression of AQP4 in NPCE and that may be one of the reasons to cause corticosteroid-induced glaucoma.
