Identification of BPI23-haFGF fusion protein and its biological activities

Abstract: Objective: To study the biological activity of BPI23-haFGF fusion protein. Methods: Recombinant expression of yeast strains pPICZαA-BPI23-haFGF/X-33 was induced by methanol. After purification by affinity chromatography and concentrated by lyophilization, the biological functions of fusion protein BPI23-haFGF with bacteria, NIH3T3 cells and mice scalded model were analyzed. Results: The purity of fusion protein BPI23-haFGF is about 90%, it has the dual function at bactericidal and mitogenetic, and the mitogenic effects of BPI23-haFGF is considerable to Standard HaFGF. It is highest (88.7%) when its concentration is 0.40 g/L. On the mice scalded model, the BPI23-haFGF group can advance healing than the negtive sample group by 2 to 3d. Conclusion: The fusion protein BPI23-haFGF can improve the healing of burn wounds, it laid the foundation for its clinical application.
