PI3K/Akt信号通路上调急性肺损伤大鼠肺泡上皮钠通道表达

摘要: 摘要:目的 探讨PI3K/Akt信号通路对急性肺损伤(ALI)大鼠肺泡上皮钠通道(ENaC)表达的影响。方法 健康成年SD大鼠随机分为对照组、ALI组(LPS)、治疗组(LPS+胰岛素)、干预组(LPS+胰岛素+渥曼青霉素),每组5只。观察肺组织病理学改变,收集支气管肺泡灌洗液(BALF),测量总肺水含量,RT-PCR法测定ENaC mRNA表达以及Western blot法测定ENaC蛋白和p-Akt表达。结果 治疗组BALF蛋白含量、髓过氧化物酶(MPO)活性、总肺水含量较ALI组显著减少(p<0.05),干预组BALF蛋白含量、MPO活性、总肺水含量较治疗组显著增加(p<0.05)。治疗组ENaC mRNA和蛋白表达及p-Akt较ALI组显著升高(p<0.05)。干预组ENaC mRNA 和蛋白表达及p-Akt较治疗组显著降低(p<0.05)。结论 激活PI3K/Akt通路通过上调ENaC表达,从而清除过多的肺水肿液。

Abstract: Abstract:Objective To investigate the role of PI3K/Akt signaling pathway on expression of alveolar epithelial sodium channel(ENaC) in LPS-induced acute lung injury (ALI). Methods Healthy adult Sprague-Dawley (SD) rats were randomly divided into control group, ALI group(LPS) ,treatment group(LPS+insulin), intervention group(LPS+insulin+wortmannin) with 5 rats in each group. The pathological changes in lung were observed, total lung water content(TLW) were measured, bronchoalveolar lavage fluid(BALF) were collected,and the mRNA and protein levels of ENaC and level of p-Akt were determined by RT-PCRand Western blot. Results Protein level and MPO activity in BALF and TLW in treatment group was significantly lower than that in ALI group (p<0.05); Protein level and MPO activity in BALF and TLW in intervention group was significantly higher than that in treatment group (p<0.05). The mRNA and protein levels of ENaC and level of p-Akt in treatment group were significantly higher than that of ALI group (p<0.05); The mRNA and protein levels of ENaC and level of p-Akt in intervention group were significantly lower than that of treatment group (p<0.05). Conclusion Activation of PI3K/Akt signaling pathway up-regulates the expression of ENaC to clear the pulmonary edema fluid.
