X线放射诱导大鼠心肌细胞系H9c2损伤中组蛋白乙酰化水平降低

Histone acetylation is decreased in X-ray radiation-induced injury of rat myocardial cell line H9c2

摘要: 目的 研究辐射暴露对心肌细胞的影响及组蛋白乙酰化的修饰变化,分析其相关性并探讨其机制。方法 将大鼠心肌细胞系H9c2分为对照组和放射组,放射组分别用X线给予2、4、6和8 Gy剂量照射。48 h后,通过比色法检测细胞培养上清乳酸脱氢酶(LDH)活性、细胞内丙二醛(MDA)含量及过氧化物歧化酶(SOD)活性;用annexin V-FITC/PI染色及流式细胞仪检测细胞凋亡率;Western blot检测凋亡相关蛋白Bcl-2、caspase3及组蛋白乙酰化相关蛋白H3K9ac、HDAC3表达。结果 与对照组相比,不同剂量照射组细胞上清LDH相对活性增加(P<0.05),细胞内MDA相对含量增加(P<0.05),细胞内SOD相对活性降低(P<0.05);此外放射组细胞凋亡率明显增高(P<0.05),并且Bcl-2蛋白表达水平降低(P<0.05),caspase-3蛋白表达水平增高(P<0.05);同时与对照组相比,放射组组蛋白H3乙酰化水平降低(P<0.05),HDAC3表达水平增高(P<0.05)。结论 辐射暴露导致H9c2细胞损伤并诱导其凋亡,此过程中细胞内组蛋白乙酰化水平降低,而HDAC3可能是调节组蛋白乙酰化水平的主要因子。

Abstract: Objective To investigate the effects of X-ray radiation on H9c2 cells and histone acetylation patterns, and to analyze the correlation between them. Methods H9c2 cells were randomly divided into control group and different model groups. The cells of model groups were irradiated with different doses of 2,4,6,8 Gy of X-ray radiation respectively. The activity of LDH in supernatants, the activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) in cells were detected by chromatometry. Cell apoptotic ratio was measured by annexin V-FITC/PI staining and flow cytometry analysis. Apoptosis-related protein Bcl-2,caspase-3 and histone acetylation related protein H3K9ac,HDAC3 were detected by Western blot. Results Compared with control group, the relative activity of LDH in supernatants and relative content of malondialdehyde (MDA) in H9c2 cells were increased (P<0.05) in model groups, the relative activity of intracellular superoxide dismutase (SOD) was decreased(P<0.05) in model groups. The apoptotic rate of H9c2 cells was increased in model groups compared with control group(P<0.05). The protein expression of caspase-3 increased (P<0.05), the expression of Bcl-2 decreased in model groups(P<0.05), and also found that the protein expression of H3K9ac significantly decreased and the expression of HDAC3 significantly increased in model groups(P<0.05). Conclusions X-ray radiation can induce H9c2 cells' injury and apoptosis,accompanied by the decrease of intracellular histone acetylation level, this change may be related to the increase of HDAC3 expression.
