α- 硫辛酸降低脂多糖诱导的Raw264.7 细胞TNF-α的分泌

α-Lipoic acid reduces TNF-α release of LPS-induced Raw264.7 cell

摘要: 目的 观察α- 硫辛酸(ALA)对脂多糖(LPS)在体外诱导的Raw264.7细胞TNF-α释放及机制。方法 用ALA或者ALA联合ERK或NF-κB的抑制剂或者激活剂预处理2 h,再用LPS(1 mg/L)体外刺激巨噬细胞Raw264.7;采用MTT法检测ALA对Raw264.7细胞活力影响,利用ELISA方法对Raw264.7释放在培养上清中的细胞因子肿瘤坏死因子(TNF-α)的浓度,及用Western blot方法检测T-pERK、pERK和NF-κB的表达。结果ALA可抑制LPS诱导的Raw264.7细胞TNF-α表达(p<0.05),用ALA联合ERK或NF-κB的激活剂预处理后可减弱ALA产生的抑制效果(p<0.05)。同时ALA抑制LPS诱导的Raw264.7细胞pERK和NF-κB的表达(p<0.05),用ALA联合ERK或NF-κB的抑制剂预处理后可加强ALA产生的抑制效果(p<0.05)。结论ALA可能通过抑制LPS诱导的Raw264.7细胞的ERK和NF-κB信号通路来抑制TNF-α释放。

Abstract: Objective To investigate the effect of α-lipoic acid on the release of TNF-α of LPS-induced Raw264.7 cell. Methods The Raw264.7 cell were pretreated with ALA or ALA and ERK /NF-κB inhibitors or activators for 2 h, and then stimulated with LPS (1 mg/L). The levels of tumor necrosis factor (TNF-α)in cultured supernatant were determined by ELISA kit.T-pERK,pERK and NF-κB were analyzed by Western blotting.Results ALA inhibited LPS-induced release of TNF-α(p<0.05),while the effect was reversible when the Raw264.7 cell were pretreated with ALA and ERK /NF-κB activators(p<0.05).ALA inhibited the phosphorytion of ERK and NF-κB (p<0.05),while the effect was strengthen when the Raw264.7 cell were pretreated with ALA and ERK /NF-κB inhibitors(p<0.05).Conclusions α-lipoic acid attenuates LPS-induced release of TNF-α by ERK and NF-κB signaling pathway.
