NF-κB抑制剂PDTC对大肠癌体外血管生成因子表达的影响

Effect of NF-κB inhibitor PDTC on the expressions of angiogenic factors in colon carcinoma in vitro

摘要: 目的 探讨NF-κB抑制剂PDTC对大肠癌Lovo细胞系体外血管生成因子表达的影响。方法 采用NF-κB抑制剂PDTC处理Lovo细胞,以未作处理Lovo细胞作为对照组,以PDTC处理后的Lovo细胞作为实验组。Western blot检测各组细胞NF-κB、p-IκBα、VEGF和b-FGF蛋白表达情况;Transwell细胞迁移实验检测HUVEC迁移能力;收集各组Lovo细胞上清液,分别加入到培养液中培养HUVEC,并采用CCK-8细胞增殖实验检测HUVEC增殖能力。结果 Western blot结果提示实验组NF-κB、p-IκBα、VEGF和b-FGF表达量均明显低于对照组(P<0.05);Transwell细胞迁移结果显示实验组HUVEC迁移穿过微孔膜的细胞数量较对照组明显减少(P<0.05),其迁移抑制率为49.2%;CCK-8细胞增殖实验结果显示实验组HUVEC增殖数量较对照组明显减少(P<0.05),其增殖抑制率为73.2%。结论 在大肠癌细胞中,NF-κB抑制剂PDTC可能通过下调NF-κB的活性而降低其体外血管生成能力。

Abstract: Objective To investigate the effect of NF-κB inhibitor PDTC on the expressions of angiogenic factors in colon carcinoma Lovo cells in vitro. Methods NF-κB inhibitor PDTC was used to treat colon carcinoma Lovo cells. Untreated Lovo cells were served as control group, Lovo cells that were treated with PDTC were served as experimental group. The expressions of NF-κB, p-IκBα, VEGF and b-FGF in each group were detected by Western blot. The ability of HUVEC migration was observed by Transwell migration assay. The supernatant of Lovo cells in each group was respectively collected and added into HUVEC culture medium separately, then CCK-8 was used to check the ability of HUVEC proliferation. Results Western blot results showed that the expressions of NF-κB, p-IκBα, VEGF and b-FGF in the experimental group were all obviously lower than that in the control group(P<0.05). The results of migration assay showed that the quantity of migratory HUVEC through the microporous membrane was decreased in the experimental group than that in the control group(P<0.05)and migratory inhibition rate was 49.2%. The results of CCK-8 showed that the proliferation of HUVEC in the experimental group was obviously lower than that in the control group(P<0.05), and the inhibition rate was 73.2%. Conclusion In Lovo cells,the NF-κB inhibitor PDTC may inhibit the ability of angiogenesis of Lovo cells in vitro through down-regulating the activity of NF-κB.
