帕金森病模型大鼠中脑黑质磷酸化α-突触核蛋白表达增加

Increased expression of phosphorylated α-synuclein in the substantia nigra of Parkinson's disease eat model

摘要: 目的 观察鱼藤酮对大鼠中脑黑质磷酸化α-突触核蛋白(α-SYN)、酪氨酸羟化酶(TH)表达的影响。方法 利用脑立体定位技术注射微量鱼藤酮,建立急性损伤帕金森病(PD)模型。采用动物行为轨迹分析软件计算各时间点30 min 内运动距离和平均运动速度。免疫组织化学检测黑质(SN)部位TH、磷酸化α-SYN的表达,并统计TH和磷酸化α-SYN阳性神经元数目。结果 PD组动物各时间点30 min内运动距离、平均运动速度低于对照组(P<0.05, 0.01)。同对照组相比,损伤侧SN部位TH阳性神经元数目明显减少(P<0.01),磷酸化α-SYN阳性神经元数目明显增加(P<0.01)。结论 PD大鼠中脑黑质磷酸化α-SYN表达增加及TH的表达明显下降,并出现运动障碍。

Abstract: Objective This experiment was to observe the effect of rotenone on phosphorylated α-synuclein and TH expression in the rat SN of PD. Methods Rats were stereotactically injected with rotenone. Movement distance and mean speed within 30-minute were calculated with Ethovison software. TH and phosphorylated α-SYN expression of the lesioned SN were demonstrated with immunohistochemical staining. The number of TH-positive neurons and phosphorylated α-SYN-positive neurons in the lesioned SN were measured. Results Movement distance and mean speed in the PD group were lower than those in the control group(P<0.05, 0.01). Compared to control group, TH-positive neurons counts in the lesioned SN in the PD group was reduced(P<0.01). The numbers of α-SYN-positive neurons were significantly higher in the PD group than in the control group(P<0.01). Conclusion Injection with rotenone could induce PD symptom, significant TH-positive neurons decrease and phosphorylated α-SYN increase.
