大鼠骨髓间充质干细胞向多巴胺样神经细胞分化

In vitro differentiation of rat bone marrow mesenchymal stem cells into dopamine-like nerve cells

摘要: 本研究目的是探索大鼠骨髓间充质干细胞向多巴胺样神经细胞分化;利用骨髓细胞贴壁生长特性,采用全骨髓培养法分离大鼠骨髓间充质干细胞;体外培养至第3代,用碱性成纤维细胞生长因子,抗坏血酸和表皮生长因子诱导分化; 用免疫荧光法鉴定胞浆中的多巴胺神经元相关蛋白的表达;RT-PCR鉴定多巴胺神经元相关基因的表达;ELISA法鉴定上清及胞浆中的多巴胺的表达;结果显示,经过诱导后,免疫荧光法可以检测到诱导后的细胞中多巴胺神经元相关蛋白:酪氨酸羟化酶、多巴胺转运蛋白和神经核蛋白;RT-PCR可以检测到诱导后的细胞表达神经细胞相关基因Nestin,Nurr-1;ELISA法可以检测到诱导后的上清及胞浆中有多巴胺;提示间充质干细胞具有向多巴胺神经细胞能力。

Abstract: To explore the differentiation of rat bone marrow mesenchymal stem cells into dopamine nerve cells. The mesenchymal stem cells were isolated and purified by cell culture base on growth characteristics of bone marrow cells. When passed third generation, they were induced to differentiate into dopamine nerve-like cells by bFGF, Vitc and EFGF. Dopamine-associated protein and genes in the treated cells was examined by immumofluorescence method and RT-PCR. Dopamine in the supernatant and cytoplasm from culture system was determined by ELISA kit. The results show that Tyrosine hydroxylase, dopamine transporter and nerve neucleprotein and Nestin,Nurr-1 genes were found. And the dopamine exists in in the supernatant and cytoplasm from inducing culture system. It is suggested rat bone marrow mesenchymal stem cells have the capacity of differente into dopamine nerve cells.
