Objective To explore the effects and mechanism of methylglyoxal (MGO) in human brain microvascular endothelial cell line (hCMEC/D3). Methods The hCMEC/D3 cells were incubated with MGO(0.25, 0.50, 0.75, 1.00 and 1.25 mmol/L) for 24 h. Cell viability was detected by CCK-8 assay. Cellular LDH and SOD activities were determined by commercially available kits. The change of mitochondrion membrane potential was assessed by JC-1 probe. MitoSOX probe was used to observe mitochondrial reactive oxygen species (mROS) release. Results Compared with control, cell viability of hCMEC/D3 treated with MGO decreased in a concentration-dependent manner (P<0.01). Increased LDH activity and decreased SOD activity were observed with MGO exposure(P<0.01). The decrease of mitochondrion membrane potential and the excessive increase of mROS were induced after MGO exposure for 12 h, which were detected by JC-1 or MitoSOX probe. Conclusions MGO reduces cell viability and mitochondria membrane potential in hCMEC/D3 cells with excessive mROS generation.

Objective To investigate the effect of sildenafil (Sil) on the expression of NLRP3/caspase-1 pathway in the aorta vascular tissue of erectile dysfunction (ED) rat models. Methods ED rat model was established and randomly divided into ED group and Sil group. Another 10 rats were selected as control group. After intragastric administration of Sil(20 mg /kg, once a day for 2 weeks), HE staining was used to observe the morphological change of aorta. Aortic fibrosis was detected by Masson staining. The content and distribution of interleukin-1β (IL-1β) and endothelial nitric oxide synthase (eNOS) in aorta were determined by immunohistochemistry. RT-qPCR and Western blot were used to detect the expression of NLRP3, caspase-1, GSDMD, IL-1β and eNOS in the aorta. Results In ED group, there were pathological changes in aortic morphology, more local exfoliation of endothelial cells and obvious fibrosis. The mRNA and protein expressions of NLRP3, caspase-1, GSDMD and IL-1β were increased, and the mRNA and protein expressions of eNOS were decreased in the aorta of the rats in the experimental group(P<0.05). Compared with the ED group, the aortic morphological changes of the Sil group were improved, the local endothelial cell shedding was reduced, and the fibrosis was alleviated. The mRNA and protein expression of NLRP3, caspase-1, GSDMD and IL-1β decreased, while the mRNA and protein expression of eNOS increased in aorta(P<0.05). Conclusions The expression of NLRP3/caspase-1 pathway protems in the aorta vescular tissue of rats with ED is up-regulated by Sil, which may improve the inflammatory response, vascular fibrosis and endothelial dysfunction by inhibiting the NLRP3/caspase-1 pathway in the aorta vascular tissue of rats with ED.

Objective To investigate the molecular mechanism of lncRNA (HOTAIR)/miR-206/TAGLN2 signaling axis in regulating the invasion of papillary thyroid carcinoma cells line TPC-1. Methods The papillary thyroid carcinoma(PTC) and paracancerous tissues were collected from the First Hospital of Lanzhou University, and the expression of lncRNA (HOTAIR), miR-206 and TAGLN2 in PTC tissues as well as paracancerous tissues was detected by RT-qPCR and Western blot; The thyroid papillary cancer cells with miR-206 over-expression and HOTAIR under-expression were constructed to detect the expression of target gene and its effect on the invasion of TPC-1 cells by Transwell assay. Results Compared with the adjacent tissue, the expression of HOTAIR in papilla-rythyroid cancer tissue was increased by 2.48 times. miR-206-5p was decreased down to 0.6,and the expression of TAGLN2 was increased (P<0.05). After over-expression of miR-206, the protein expression of TAGLN2 was decreased in TPC-1 cell, and the invasion of TPC-1 cells was inhibited (P<0.05). Knockdown of HOTAIR increased expression of miR-206 in TPC-1 cells and decreased the expression of TAGLN2. The Transwell assay showed that the invasion of TPC-1 cells was significantly reduced (P<0.05). Conclusions In PTC, reducing the expression of HOTAIR results in over-expression of miR-206 then further inhibits the expression of TAGLN2. The biological outcome is inhibition of invasion of thyroid cancer.

Objective To investigate the effect of gllic acid (GA) on the pulmonary inflammatory response associated with chronic obstructive pulmonary disease (COPD) induced by cigarette smoking (CS) in mice. Methods The mouse model induced by CS was constructed. The counting number of inflammatory cells and the level of proinflammatory cytokines in bronchoalveolar lavage fluid were analyzed. The ROS, GSH, MDA and protein carbonyl activity in lung tissue homogenate were determined by commercially available kits. Western blot was used to analyze IL-13/STAT6 pathway. Results GA inhibited SC-induced inflammatory cell infiltration (neutrophils, lymphocytes, and macrophages) and decreased the level of pro-inflammatory cytokines (IL-6, TNF-α, and IL-1β). GA alleviated the redox imbalance and CS-induced airway inflammation in lung tissue of CS exposed mice. The later might be mediated by by inhibiting IL-13/STAT6 signaling pathway. Conclusions GA effectively modulates pulmonary inflammation and oxidative stress in mice associated with COPD pathogenesis.

Objective To investigate the detection and clinical application studies based on malaria protein VAR2CSA(VAR2) synthetic peptide specifically binding to circulating tumor cells (CTCs). Methods The peptide sequence of the functional structural domain of malaria protein VAR2CSA was selected to synthesize VAR2 that enriched CTCs based on nano-microfluidic technology, and used VAR2 synthetic peptide to detect enriched cells to establish a novel detection protocol for CTCs. Cellular immuno-fluorescence staining, flow cytometry and tumor cell doping recovery assays were used to analyze the specific binding and recovery efficiency of VAR2 synthetic peptide with tumor cells of different tissue sources and compare with immuno-staining-fluorescence in situ hybridization (imFISH) assay. Twenty-two colorectal cancer patients and twenty-two healthy individuals were selected, and the VAR2 synthetic peptide was used to detect peripheral blood CTCs in both. The VAR2 synthetic peptide was used to compare the positive detection rate of CTCs in patients with different TNM stages. Results The VAR2 synthetic peptide bound specifically to tumor cells collected from different tissue sources of human colon cancer cell line SW620, human breast cancer cell line MCF7 and human esophageal squamous carcinoma cell line KYSE180.The VAR2 synthetic peptide detected 100 SW620, MCF7 and KYSE180 tumor cells mixed into 3 mL of peripheral blood with recovery rates of 82.8%±8.41%, 56.2%±3.57%, 86.6%±8.19%, which were not significantly different from the imFISH assay. The VAR2 synthetic peptide assay recoveries of 50, 100, and 200 SW620 tumor cells were 80.7%, 82.8%, and 84.7%, respectively, which did not correlate significantly with the number of adulterated tumor cells. The VAR2 synthetic peptide detected 63.6% positive CTCs in peripheral blood samples from patients with clinical colorectal cancer, which was higher than the 0.0% positive CTCs rate in the group of healthy subjects, and also correlated with depth of invasion, lymph node metastasis, distant metastasis, and clinical stage (P<0.05). Conclusions The VAR2-based synthetic peptide may be applied in specific and non-antibody dependent CTCs detection in different tissue samples and so has good clinical application value for clinical staging and severity evaluation of tumor patients.

Objective To explore the mechanism of action of urantide in ameliorating renal injury caused by atherosclerosis(AS). Methods The rats were divided into normal group, model group which were treated with ip VitD3 150 U/(kg·d-1) for 3 consecutive days, and fed with high-fat special diet, with an experimental period of 6 weeks], simvastatin group [5 μg/(kg·d-1) for 14 consecutive days] and intervention model group [urantide 30 μg/(kg·d-1),7 d and 14 d]. Microscopy was performed for observing pathological changes in the kidney and evaluation of the degree of renal tissue damage. RT-qPCR and immuno-histochemical staining microscopy was used to detect the expression of related genes and proteins. Results Compared to the normal group, the model group showed glomerular atrophy in the renal tissue and adhesion with surrounding tissues. The experiment found a series of changes in renal tubular epithelial cells, such as edema and degeneration, accompanied by a small area of cell necrosis. Sclerosis was found in the glomerulus; Increased positive expression of genes and proteins related to JAK2 and STAT3 (P<0.05,P<0.01)was found . Compared to the model group, urantide groups with prolongated medicine administration and the pathological changes that occur in the kidney tissue showed obvious improvement and upturned. At the same time, the degree of glomerulosclerosis decreased and renal tubulointerstitial injury was improved(P<0.01); Positive expression of genes and proteins related to JAK2 and STAT3 (P<0.05, P<0.01) all decreased. Conclusions Urantide may inhibit JAK2/STAT3 signal pathway and this finding may support the development of clinical strategy for the prevention and treatment of AS related renal injury.

Objective To investigate the impacts of pachymic acid (PA) on proliferation and apoptosis of human colon cancer cell line (HT-29) and its potential mechanism. Methods The cells were divided into blank group, 20, 40 and 80 μmol/L PA groups. The proliferation of HT-29 cells was detected by MTT method; The cell cycle and apoptosis of HT-29 cells were detected by flow cytometry; The expression of cell cycle, apoptosis and Akt/MDM2/p53 signal pathway related proteins in HT-29 cells were detected by Western blot. The tumor xeno-transplantation model with HT-29 cells was constructed, and then the tumorigenic nude mice were randomly divided into control group, intra-peritoneal injection of 10, 30 and 60 mg/kg PA groups(6 in each group), the tumor volume and weight were measured; The expression of Ki-67 and of cleared caspase-3 proteins in tumor tissue was detected by immuno-histochemistry; The expression of Akt/MDM2/p53 signal pathway related protein in tumor tissue was detected by Western blot. Results PA of all doses obviously inhibited proliferation of HT-29 cells, the proportion of cells in S phase and in G2/M phase was deceased and the expression of B cell lymphoma gene-2(Bcl-2), cyclin D1, cycle protein dependent activating enzyme 2(CDK2) proteins all decreased, The ratios of p-Akt/Akt and p-MDM2/MDM2 in a concentration-dependent manner increased(P<0.05).The apoptosis rate, the ratio of cells in G0/G1 phase and the protein expression of Bax, cleaved caspase-3 and p53 also increased(P<0.05). PA in all dose groups obviously reduced the tumor volume, weight, the expression of Ki-67 protein and the ratios of p-Akt/Akt, p-MDM2/MDM2 in tumor tissue of nude mice (P<0.05), and increased the protein expression of cleaved caspase-3 and p53 in tumor tissue(P<0.05). Conclusions PA can induce apoptosis and cell cycle arrest of colon cancer cells and inhibit cell proliferation. Its mechanism may be related to the regulation of Akt/MDM2/p53 signal pathway.

Objective To screen the potential key genes associated with colon cancer(CC) by bioinformatics analysis based on Gene Expression Omnibus (GEO) database, and verify them through in vitro experiments to explore potential colon cancer molecular markers. Methods The human CC data sets GSE10950 and GSE74602 were obtained from GEO database. The differentially expressed genes (DEGs) in colon cancer and adjacent tissues were screened by GEO2R tool and Venn diagram software. DEGs were used to perform by Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). The Protein-Protein Interaction (PPI) network of DEGs was constructed by STRING database, then the key genes were screened with Cytoscape software. GEPIA database was used to validate expression and prognostic value of key genes. Finally, the functions of key genes were further verified by cell transfection and CCK8 assays. Results 515 DEGs were obtained from two datasets, of which 223 were up-regulated and 292 were down-regulated. GO enrichment analysis showed that up-regulated DEGs were mainly involved in negative regulation of cell cycle, transcriptional control. KEGG signaling pathways analysis showed that up-regulated DEGs were mainly enriched in cell cycle signaling pathways. A total of 33 hub genes were screened through the PPI. UCLCAN analysis was implemented and 7 of 33 genes were relative to lower survival rate of CC patients. The expression levels of 7 genes by GEPIA analysis (P<0.05).It was verified by GEPIA database that CCNB1,CCNA2,CDC20,CDKN3,DLGAP5,HMMR and NCAPG were highly expressed in colon cancer tissues. KEGG pathway enrichment and found that three critical genes (CCNB1, CDC20 and CCNA2) enriched in cell cycle pathway. The inhibition of CCNB1,CDC20 or CCNA2 significantly promoted proliferation of colon cancer cells. Conclusions A total of 7 key genes were identified to be involved in the occurrence of CC. The expression of CCNB1,CDC20 and CCNA2 is potentially related to the poor prognosis in CC, which may be potential targets of clinical treatment and prognostic markers for CC patients.

Graft-versus-host disease (GVHD) is one of the most serious complications after allogeneic hematopoietic stem cell transplantation (allo-HSCT), resulting a very high mortality risk and no effective diagnosis and treatment method. Although an increasing number of studies suggest that immune-mediated vascular endothelial damage plays an important role in GVHD, the specific mechanism remains elusive. On the basis of endothelial injury caused by radiotherapy and chemotherapy before allo-HSCT and infection, the donor T cells directly contact with the damaged endothelial cell antigen, secrete cytokines and inflammatory mediators to kill endothelial cells, leading to further apoptosis and depletion of endothelial cells, and exacerbating the functional damage of target organs and multi-system injury of the body. To explore the endothelial injury mechanism of GVHD. This review provides a new idea for the prevention and treatment of GVHD.

Calcium binding and coiled-coil domain 1 (CALCOCO1) is an autophagy regulatory protein being discovered in recent years, which plays an important role in selective autophagy process. The gene coding for autophagy is located at 12q13.13, and its coding protein is CALCOCO1 protein. Which functions by binding to ATG8 family proteins, it anchors to the endoplasmic reticulum and the Golgi apparatus and thus initiate the autophagy of endoplasmic reticulum and Golgi aparatus, and thereby participating in selective autophagy and early stages of autophagosome formation. When the mTOR pathway is activated, mTOR regulates the expression of CALCOCO1 through the autophagy-dependent pathway. CALCOCO1 plays a role in the occurrence and metastasis, and so is a potential target for chemotherapy of malignant tumors.

Objective To explore the application effect of micro-lecture combined with case-based learning (CBL) in the teaching of pharmacotherapeutics for undergraduate nursing students. Methods Two classes of undergraduate nursing students from School of Nursing, Peking Union Medical College enrolled in 2021 were selected as the research subjects. They were taught by micro-lecture combined with CBL (experimental group) or traditional lecture-based learning (LBL) (control group) respectively; The students' in-class quiz and final exam scores were collected. The questionnaires were distributed to evaluate the teaching effect and learning experience through analyzing the scores and questionnaire results. Results The in-class quiz scores of the experimental group were 77.52±17.68, and those of the control group were 66.34±21.35; The final exam scores of the experimental group were 64.34±26.17, and those of the control group were 52.33±25.55. The scores of the experimental group in both tests were significantly higher than those of the control group, and the differences were statistically significant (P<0.01). The questionnaire findings indicated that the experimental group improved on the theoretical understanding, knowledge retention, medication practice proficiency, interest in learning, concentration in class, and interaction willingness in comparison to the control group. Conclusions Micro-lecture combined with CBL can enhance the learning effect of pharmacotherapeutics for undergraduate nursing students, and optimize the learning experience. It is worth to be further improved and promoted as a feasible teaching method.

The ultrasound diagnosis imaging report reflects the sum of a radiologist's synthesis and judgement into a patient's condition. It is also an important product that is generated by radiologists to help the patients. In spite of the importance of clear and effective report writing, radiologists usually receive little or no formal reporting education. There are key principles when reporting the ultrasound findings, impression, and recommendations to promote report quality and reading experience. The findings should emphasize short and factual observations while avoiding redundancy. The impression is the thoughtful synthesis of findings leading to a diagnosis, a differential diagnosis and management recommendations. Improved report writing is achieved only after persistent learning, follow up and summaries.
