地塞米松抑制兔眼睫状体无色素上皮水通道-4蛋白的表达

Dexamethasone inhibits expression of aquaporin-4 of non-pigmented ciliary epithelium in rabbit

摘要: 目的 研究地塞米松磷酸钠对兔眼睫状体无色素上皮(NPCE)水通道蛋白-4(AQP4)表达的影响。方法 建立兔皮质类固醇性高眼压模型,用免疫组化、免疫荧光和western blot法检测兔眼AQP4表达水平。结果 免疫组化检测显示高眼压组AQP4的表达水平(A=0.119±0.072)比正常对照组(0.242±0.056)下降(p<0.05),停药后眼压和病理改变也逐渐恢复(0.119±0.072), Western blot结果表明高眼压组AQP4的表达强度(IA=63.54)比正常对照组的(167.00)明显减弱(p<0.05)。结论 地塞米松磷酸钠诱抑制兔眼NPCE上AQP4的表达,增加睫状体房水的分泌,这可能是皮质类固醇性青光眼的病理机制之一。

Abstract: Objective To study the effects of dexamethasone (Dex) on the expression of aquaporin-4(AQP4) in rabbit non-pigmented ciliary epithelium(NPCE). Methods Firstly to construct rabbit corticosteroid-induced glaucoma model and then contrast the differences of AQP4 expression in NPCE with normal controls by immunohistochemistry and immunofluorescence methods, as well as western blot method. Results The immunohistochemistry tests indicated that the AQP4 expression in NPCE of DEX group (OD=0.119±0.072) was suppressed than normal controls (0.242±0.056) (p<0.05). After some time the IOP decreased some of above changes gradually recovered (0.119±0.072), so did the western blot tests. Conclusions DEX increased IOP by changing the expression of AQP4 in NPCE and that may be one of the reasons to cause corticosteroid-induced glaucoma.
