摘要: 自然杀伤细胞(NK)是一种重要的先天性免疫细胞,无需预先抗原致敏,能够直接识别和攻击病毒感染的细胞和肿瘤细胞。NK细胞的活化和效应功能受NK细胞表面的激活受体和抑制受体传递的信号动态平衡调控,通过与靶细胞表面的配体结合,依据“诱导自我”、“缺失自我”模式实现细胞毒性作用。NK细胞的主要杀伤机制包括:释放穿孔素和颗粒酶等细胞毒性颗粒诱导靶细胞裂解;死亡受体途径介导的细胞凋亡;分泌多种细胞因子、趋化分子和生长因子,协同其他免疫细胞杀伤肿瘤细胞,从而产生次级免疫应答;抗体依赖细胞介导的细胞毒性作用。

Abstract: Natural killer cells (NK) are important innate immune cells that do not require prior antigen exposure and can directly recognize and attack virus-infected cells and tumor cells. The activation and effector functions of NK cells are regulated by a balance of signals delivered through their surface activating receptors and inhibitory receptors, which bind to ligands on target cells to achieve cytotoxicity via “induced self” and “missing self” recognition models. The killing mechanisms of NK cells primarily include release of cytotoxic granules such as perforin and granzymes to induce target cell lysis, death receptor-mediated apoptosis, secretion of various cytokines,chemokines and growth factors to coordinate with other immune cells in killing tumor cells, thereby generating secondary immune responses and antibody-dependent cellular cytotoxicity(ADCC).
