人胚胎干细胞抑制人肝癌细胞系SK-Hep1增殖并促进凋亡

Human embryonic stem cells inhibits the proliferation and promote apoptosis of SK-Hep human hepatoma cell line SK-Hep1

摘要: 目的 探究在人胚胎干细胞hESCs与人肝癌细胞系SK-Hep1共培养微环境中,hESCs对SK-Hep1人肝癌细胞增殖、侵袭、迁移和凋亡的影响。方法 建立hESCs与SK-Hep1人肝癌细胞系的非接触式共培养体系,将与hESCs共培养的SK-Hep1细胞作为实验组,单独培养的SK-Hep1细胞作为对照组。MTT法检测SK-Hep1细胞的增殖能力;Transwell小室法检测SK-Hep1的侵袭与迁移能力;Hoechst33258染色后观察共培养后SK-Hep1细胞核的变化;流式细胞术检测SK-Hep1细胞凋亡率。结果 与hESCs共培养后,SK-Hep1细胞的增殖能力受到抑制,随着时间增加,抑制效果越显著(P <0.05);细胞侵袭、迁移穿过Transwell小室的数量显著减少(P <0.05);发生核固缩、变形和浓染的SK-Hep1细胞与对照组比增多;细胞凋亡比率与对照组比显著增加(P <0.05)。结论 人胚胎干细胞对SK-Hep1人肝癌细胞系有抑制作用。

Abstract: Objective To explore effects of human embryonic stem cells ( hESCs ) onhproliferation , invasion and migration abilities of SK-Hep1 humanhepatoma cells in the co-culture micro environmen of hESCs and SK - Hep1. Methods Single cultured SK-Hep1 cells were served as control group while SK-Hep1 which non-contact co-cultured with hESCs was regarded as experimental group.The hproliferation ability of SK-Hep1 was measured by MTT method; invasion and migration ability of SK-Hep1 cells were detected by Transwell chamber method ; the nucleus variation and cell apoptosis of SK-Hep1 were detected by Hoechst33258 chromosome and flow cytometry. ResultsThe proliferation ability of SK-Hep1 cells in the experimental group was inhibited obviously compared with control group (P <0.05); the number of SK - Hep1 cells which passed through the Transwell chambers were reduced significantly compared with control group in invasion and migration experiment(P<0.05); more nucleus pycnosis and deformation appeared in experimental group than that in control group .And apoptosis rate of SK- Hep1 cells in the experimental group was significantly higher than that of in the control group (P <0.05). Conclusions Human embryonic stem cells have inhibitory effect on SK-Hep1 human hepatoma cell line.
