熊果酸对THP-1细胞HMGB1表达和NF-кB活性的影响

Effects of ursolicacid on HMGB1 expression and NF-кB activity of THP-1 monocytes

摘要: 目的 研究熊果酸(UA)对人单核细胞系THP-1细胞HMGB1表达和NF-кB活性的影响。方法 用不同浓度的UA(0、0.1、1、5和10μmol/L)作用细胞后进行如下检测:1)MTT检测细胞增殖;2)RT-PCR法检测HMGB1和p65 mRNA表达;3)Western blot检测HMGB1和p65蛋白的表达;4)荧光素酶报告基因转染,质粒pNF-κB-luc瞬时转染THP-1细胞,转染40h后加1μmol/LUA作用,于转染48h后进行荧光素酶活性测定。结果 1)随着UA浓度的增高,可明显抑制THP-1细胞的增殖;2)不同浓度的UA对THP-1细胞HMGB1 mRNA和蛋白表达,呈现出一定的双向性,且以1 μmol/L UA作用最强(P<0.05);3)UA可增强p65 mRNA和蛋白表达以及NF-κB活性。结论 UA可影响单核细胞HMGB1表达和NF-кB活性。

Abstract: Objective To investigate the effects of ursolic acid on the expression of HMGB1 and activity of NF-κB in THP-1 cells. Methods After being stimulated by different concentrations UA (0,0.1,1,5 and 10μmol/L), the cells were detected correspondingly.⑴The cell proliferation were checked by MTT. ⑵The HMGB1 and p65 destination genes were amplificated with PCR.⑶HMGB1 and p65 protein was detected by Western blot. ⑷Transfecting of Plasmid pGl3-NF-кB-luciferase report gene:Plasmid pGl3-NF-кB-luciferase report gene was transient transfected into THP-1 cells.After transfecting for 40 hours, 1 μmol/L UA were added. Luciferase activity detection was performed when transfecting for 48 hours. Results ⑴UA can obviously inhibit THP-1 cell proliferation with the concentration increasing.⑵The influence of UA on THP-1 cell HMGB1 mRNA and protein exhibits a certain bidirectional and the influence of 1 μmol/L UA is the strongest.⑶UA can influence the expression of p65 mRNA and protein andaffect the NF-кBactivity.Conclusion UA can influence the HMGB1 expression and NF-кB activity of THP-1 cell.
