阿魏酸通过雌激素受体α调控人乳腺癌细胞系的增殖、迁移及侵袭

Ferulic acid regulates the proliferation, migration and invasion through estrogen receptor α in human breast cancer cell line

摘要: 目的 观察阿魏酸是否调控人乳腺癌细胞系MCF-7长非编码RNA(lncRNAs)的表达水平及对细胞增殖,迁移及侵袭的影响。方法 0,1、2.5、5、7.5或10 mmol/L阿魏酸处理24 h后,CCK-8法检测细胞活力(n=3);软琼脂成瘤实验检测细胞体外成瘤能力;0或2.5mmol/L阿魏酸处理24h后,划痕实验及Transwell穿膜实验检测细胞增殖、迁移及侵袭能力(n=3);基因干扰和过表达技术检测雌激素受体α(ERα)是否参与阿魏酸对乳腺癌细胞的调控;反转录-实时荧光定量PCR(RT-qPCR)检测MALAT-1 mRNA表达。结果相比于对照组,阿魏酸呈浓度依赖性增加MCF-7细胞活力增殖、成瘤、迁移和侵袭能力(P<0.05)。干扰ERα减弱阿魏酸对MCF-7细胞生理过程的调控;过表达ERα增强阿魏酸对MB231细胞的调控。同时发现阿魏酸通过ERα调控MALAT-1的表达。结论 阿魏酸通过MCF-7中ERα的介导,调控了MALAT-1的表达与乳腺癌细胞的增殖,迁移及侵袭。

Abstract: Objective To figure out whether ferulic acid regulates the proliferation, migration and invasion in human breast cancer cell line MCF-7. Methods After 0, 1, 2.5, 5, 7.5 or 10 mmol/L ferulic acid treatment for 24 h, CCK-8 assay was performed to detect cell viability and colony formation was detected by soft agar assay. After 0 or 2.5 mmol/L ferulic acid treatment for 24 h, scratch assay and transwell assay were performed to reveal migrating and invasing abilities. Gene expressing silencing and overexpression were employed to evaluate whether estrogen receptor α was involved in the regulatory role of ferulic acid. RT-qPCR analysis was performed to detect MALAT-1 levels after ferulic acid treatment. Results With the growing concentration of ferulic acid, cell viability was promoted (P<0.05). Compared to control group, ferulic acid treatment promoted proliferation, colony formation, migration and invasion in MCF-7 in a concentration dependent manner (P<0.05). Silencing of ERα decreased the effect of Ferulic acid on MCF-7; overexpression of ERα increased the effect of ferulic acid on MB231. It was also revealed that ferulic acid exposure regulated MALAT-1 expressing level in a ERα dependent manner. Conclusion Ferulic acid serum regulates lncRNA MALAT-1 and the proliferation, migration and invasionin MCF-7 cells depending on the existence of ERα receptor.
