    Treatment of Parkinson's disease-like rat model by neural stem cell transplantation

    Objective: To investigate the survival, differentiation and action of neural stem cell transplanted into striatum of Parkinson's disease rat model. Methods: Neural stem cells were separated and cultured in vitro. The character of self-renewing was confirmed by technique of single cell clone culture. The ability of multi-directional differentiation and specific antigen-nestin was showed by immunocytochemistry(ICC). The models of PD rat were established, and the DAPI-labelled neural stem cells were transplanted into striatum. The 6-OHDA induced rotary behavior was measured, distribution and migration of the cell were checked by fluoresce activating. ICC and HPLC are used to see its differentiation and action of secrete neurotransmitter dopamine(DA) and it's metabolites. Result: the cultured stem cell express nestin and have the ability of self-renewing and multi-directional differentiation. 6-OHDA induced rotation decreased after transplantation of NSC into the PD rat striatum(P<0.05)。NSC migrate widely in the recipient and form small member of TH cell, fibre in striatum. Content of DA and its metabolite (dopac) of rat's striatum after transplanting increased compare with the pseudo-transplanted group. Conclusion: transplanted neural stem cell can survived in the brain of PD rat and differentiate to cells suit to microcircumstances by excreting neurotransmitter and improve the symptom of PD.

    Objective The aim of this study was to explore the effects of SLC concentration gradient on suppression of tumor immune escape. Methods According to different SLC concentration, there were six groups. The HLA-Ⅰexpression and apoptosis of MCF-7 were detected with FCM ,and intracellular Bcl-2 expression was analyzed by western blot. The productions of TGF-βwere detected with ELISA. Results In a certain range of concentration gradient, following SLC increase, HLA-Ⅰexpression level on MCF-7 was improved, and the apoptosis was induced but Bcl-2 expression was enhanced ,moreover, the secretion of TGF-β was suppressed. Conclusion SLC inhibited tumor immune escape.

    ABSTRACT:Objective To detect the expression level of RKIP in human lung carcinoma cell lines and investigate the relationships between RKIP and cell metastasis. Methods The expression level of RKIP in human lung carcinoma cell lines was analyzed by RT-PCR. Construct and express human RKIP fusion protein and immunize New Zealand rabbit with the purified fusion protein to prepare polyclonal antiserum.The antiserum titer was identified by ELISA. The specifcity of antiserum and expression level of RKIP in human lung carcinoma cell lines was analyzed by western blotting. Results The fusion protein was expressed successfully. The prepared polyclonal antibody reacted specifically with RKIP in human cells. Downregulation of RKIP was detected in three lung carcinoma cell lines with metastasis, and it is more significant in A549 with higher metastasis. Conclusion The RKIP antiserum with high titer and good specificity could satisfy the requirement for studying on RKIP. Downregulation of RKIP has correlation with human lung carcinoma cell metastasis.

    Objective To study the cell killing effect on isolated sarcoma 180 cells by ultrasound activating Protoporphyrin IX and to explore its potential biological mechanism .Methods The sonodynamical effect was investigated on S180 tumor cells exposed to the combination of 120μmol/L protoporphyrin IX (PPIX) and focused ultrasound at the frequency of 2.2MHZ and an intensity of 3W/cm2. The livability of cells was evaluated by trypan blue staining method. Scanning electron microscope (SEM) observation of the surface of cells was performed to evaluate the morphological changes induced by ultrasonic irradiation. The generation of oxygen free radicals in cell suspensions was immediately detected after treatment by the active oxygen detection kit. Oxidative stress was assessed by measuring the activities of key antioxidant enzymes (ie, Superoxide dismutase[SOD], Glutathione peroxidase [GSH-PX], Catalase [CAT]) in S180 cells after SDT. Results The cell damage rate of ultrasound combined with PPIX was significantly higher than treatment with ultrasound alone only, and PPIX alone had no killing effect on S180 cells. Enzymatic chemical methods showed the content of MDA significantly increased after treatment, while the activities of key antioxidant enzymes in tumor cells all decreased at different levels, and which were associated to the generation of oxygen free radicals in cell suspension after treatment. Conclusion Oxygen free radical may play an important role in improving the membrane lipid peroxidation, decreasing the activities of key antioxidant enzymes in cells, and the biological mechanism might be involved in mediating the killing effect of S180 cells in SDT.

    Objective To investigate the roles of P53/P21 during neuronal differentiation with a differentiated model of PC12 cells. Methods A new cell line PC12(P53/m175) was created by stable transfection of a retrovirus plasmid pBabe-P53/m175, which contains a dominant-negative P53 gene mutant. After NGF treatment, observing with phase-contrast microscopy, flow cytometric analysis and western blotting of P53 and P21 were performed to compare with the control group. Results Expression of P53 and P21 was obviously increased in NGF-induced PC12 cells. The appearance of cell cycle G1 phase arrest paralleled the increased expressions of P53 and P21. The level of P21 protein did not change after treatment with NGF in PC12(P53/m175) cells and the extent of G1 phase arrest markedly decreased by contrast with the control group. However, we could observe the normal neurite outgrowth in NGF-treated PC12(P53/m175) cells. Conclusion NGF-induced the increased protein levels of P53 and its transcriptional element, P21 is essential for cell cycle G1 phase arrest, but does not necessarily exclusively correlate with the neurite outgrowth of PC12 cells.

    Objective:To observe the protective effects of apelin-13 on on the expression of APJ (putative receptor protein related to the angiotensin receptor AT1)and Akt1/2 in ischemia/reperfusion injury rat heart in virtro Methods Langendorff device was used to perfuse isolated rat heart with constant flow. The ischemia/reperfusion models of rat heart in vitro were made through stopping and reperfusion of the KH fluid to the heart;observing the change of maximal rate of the pressure increase(+dp/dtmax) and maximal rate of the pressure decrease(-dp/dtmax) of left ventricle in rat heart . RT-PCR were used to assay the expression of APJmRNA in rat heart, Western blotting were used to assay Akt1/2 protein in rat heart. Results The groups given apelin-13 had higher ±LVdp /dtmax(p<0.01); and resulted in hyper-expression of Akt1/2 protein (p<0.05), while the expression of APJmRNA were up regulated in IR group(p<0.05). Conclusion: Apelin-13 can resist the functional impairment in rat heart caused by ischemia/reperfusion injury, and the protecting effects of apelin-13 may involved in up regulated expression of APJ in rat heart, and the APJ might cause up regulated expression of Akt1/2 which would do good to I/R injury heart.

    Objective To investigate the effect of intracerebroventricular injection of morphine and Orphanin FQ (OFQ) on somatosensory evoked potential (SEP) and Na+-K+ATPase activity in cerebral cortex of rat. Methods Study the SEP with the BL-420 biological signal collecting system. The cerebral cortex tissues were extracted, homogenized and centrifuged. Na+-K+ATPase activity was measured with ATPase analytical box. Results After intracerebroventricular injection with OFQ 0.9μg, the amplitude of P1-N1 and N1-P2 increased significantly (P<0.01). After intracerebroventricular injection with OFQ (0.04, 0.45, 0.9, 1.8 μg), the Na+-K+ATPase activities were significantly decreased, with the increase of concentration of OFQ (P<0.01). After intracerebroventricular injection with morphine (20μg), the amplitude of P1-N1 and N1-P2 decreased significantly and the activity of Na+-K+ATPase increased significantly (P<0.01). After intracerebroventricular injection with OFQ (0.9μg) and morphine(20μg), the amplitude of SEP and the Na+-K+ATPase activity were not significantly different as compared with control groups. Conclusion It suggests that the effect of OFQ is related to the changes of the SEP amplitude and the Na+-K+ATPase activity in CNS.

    objective To study the effect of nerve growth factor(NGF) on secretion of Th1/Th2 cytokines. Methods (1)32 SD rats were divided into 4 groups randomly: asthma(the asthmatic models were established), control, NGF, anti-NGF-antibody groups. After 14 days the lung tissues were detected the pathologic changes by HE staining. NGF in the lungs were determined by enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry assay。The levels of interferon-γ(IFN-γ, one of the Th1 cytokines) and interleukin-4 (IL-4, one of the Th2 cytokines) were detected by ELISA. Results In the asthma group, more inflammatory cells were detected in the lungs. more NGF-immunoreactive infiltrating cells and higher levels of NGF(P<0.001) in the lungs were detected. The levels of IL-4 (P<0.05)were enhanced and the levels of IFN-γ(p<0.001)were decreased significantly in the lungs. In the lungs of the asthma group, the significant negative correlation was found between NGF and the ratio of IFN-γand IL-4(P<0.01). In the NGF group, the changes were enhanced significantly than those in the asthma group. on the contrary, in the the anti-NGF-antibody group, the changes were decreased significantly than those in the asthma group. Conclusion In addition to inducing neurogenic inflammation in asthmatic rats, NGF may contribute to promote amplify Th1/Th2 cells immune imbalance in asthma by enhancing Th2 cytokines and decreasing Th1 cytokines.
    Chondroitin Sulfate's Effect on the Proliferation of HL60 Cells

    objective To determine the proliferative activity of HL60 cells affected by chondroitin sulfate (ChS), to inquire the ChS's application value in chemotherapy of malignant tumor. methods The proliferative activity was measured using cell count after added ChS with different concentration. Add ADR to each group of ChS with different concentration , compare the different surviving rate of cell with other groups.The acid phosphatase activity of each group was examined with ELISA in order to observe the differentiation of HL-60 cell. results The concentration of ChS(<50mg/l) can increase the cell density of HL60 obviously (P<0.01), but the density of cell showed a little difference when the concentration increased above 50 mg/l (P>0.05). After ADR had been added, the cell's surviving rate of each ChS group showed a decrease in different degree, especially the ChS concentration above 25 mg/l (P<0.01).The acid phosphatase activity of each group increased very obviously,especially in 75mg/l group. conclusion ChS with a proper concentration can accelerate the proliferative activity of HL60 cells; And it can increase the sensibility of HL60 cell which added ADR and increase the killing effect of ADR obviously to HL60 cell.

    Objective To study the apoptosis of human hepatocarcinoma cells induced by genistein and its mechanism. Methods Human hepatocarcinoma SMMC-7721 cells cultured in vitro were treated with genistein. MTT test was used to examine the proliferation inhibitory rate, The morphology of apoptotic cells was observed by microscopy. DNA fragmentation was examined by gel electrophoresis. The expressions of apoptotic gene of P53、SURVIVIN and CASPASE-3 were examined by immunohistochemstry method. Results Genistein could inhibit the proliferation of human hepatoma cells in dose and time-dependent manner, The inhibitory rates were 9.86%、19.13% and 25.64% differently in 5,10 and 20μg /mL for 48h. It also could induce the apoptosis of SMMC-7721 cells, The DNA of cells presented "ladder" break. The expression of apoptosis related protein SURVIVIN was down regulated, the expressions of P53 and CASPASE-3 were up regulated. Conclusions Genistein can induce the apoptosis of SMMC-7721 cells. One of the molecular mechanisms of genistein might decrease apoptosis related protein expression of SURVIVIN and increase the expressions of P53 and CASPASE-3.
    Inhibitory effect of ghrelin on adipocytic differentiation

    objective: To investigate the effect of ghrelin on adipocytic differentiation of bone marrow stromal cells (BMSCs) .Methods: BMSCs from mice were cultured , and adipocytic differentiation was induced by MDI inducers .The morphological changes of the cells were observed .Oil-Red O staining measurement , alkaline phosphatase (ALP) activity measurement ,MTT assay ,RT-PCR and Western blot were employed . Results: 10-9,10 -8,10-7 mol/L ghrelin could dose-dependently inhibit the adipocytic differentiation of BMSCs and promote ALP activity (P<0.05 or P <0.01). Ghrelin decreased the expression of PPARγ2, adipsin mRNA and PPARγ2 protein. Ghrelin exerted no influences on clonal expansion in early phase of differentiation. Conclusion:ghrelin can inhibit the adipocytic differentiation of BMSCs ,which might be closely related to the downregulation of PPARγ2.

    Objective To explore the fuction of polyvinylpyrrolidone(PVP) and Adriamycin(ADM) on human bladder cancer cell line T24 and its effects of preventing recurrence of superficial bladder cancer. Methods Cell Suppression rates were treated by MTT assay. Adhesive attraction and cell cycle were measured by flow cytometry. 231 postoperative cases with superficial bladder cancer received intravesical instillation with ADM combined with PVP (experiencial group) or NS (control group). Results Cell Suppression rates were 15.11% and 49.57%, when treated with 2.5% PVP for 24 and 72h respectively; the rates increased to 35.42% and 79.66%, when treated with 7.5%PVP. Cell Suppression rate for ADM alone was 39.05%, while co-treated with 2.5% and 7.5%PVP for 48h, the rates increased to 68.51% and 88.39% respectively. 5%PVP significantly blocked the cell cycle progression, decreased G0/G1 phase and increased G2/M phase. Adhesive attraction to T24 cell was significantly increased when treated with 5%PVP. 194 cases were followed up. Compared with control group recurrence rate was clearly decreased (60.0% vs 18.6%, P<0.01) and recurrence time was remarkly delayed (58.8m vs 20.8m, P<0.01) in experiencial group. Conclusions PVP could inhibit human bladder cancer T24 cell growth by inducing cell cycle arrest at G2/M phase and improving adhesive action, the effect could be increased by co-treated with ADM. Intravesical instillation of PVP and ADM is effective for preventing tumor recurrence with fewer side effects.

    Objective To investigate the effects of extracelluar signal regulated kinase(ERK) pathway on the expression of Smad2/3 protein and mRNA in human umbilical artery smooth muscle cells(hUASMC). Methods hUASMC were primary cultured in vitro,then the hUASMC were divided into four groups:①control group,②PDGF group,③ERK blocking agent group,④PDGF+ ERK blocking agent group.The expression of phosphorylated smad2/3(p-smad2/3) protein in hUSMC were detected by inmunohistochemistry method and the expression of smad2/3 mRNA were detected by RT-PCR technology.Result ①The expression of p-smad2/3 protein in hUSMC of PDGF group was higher than that of control(p<0.01); ②There was no difference on the expression of smad2/3 mRNA in hUSMC between the four groups.Conclusion The ERK pathway could promote the expression of p-smad2/3 protein and have no effection of the expression of smad2/3 mRNA in hUASMC.
    Losartan improved heart function and decreased serum uric acid content in chronic heart failure patients

    Objective To investigate the efficacy of losartan on heart function and uric acid in patients with chronic heart failure (CHF). Methods 85 patients with CHF were divided into two groups randomly. The routine anti-CHF treatment included cardiotonic, diuresis, isosorbide dinitrate, beta-blockers. In losartan group(n=43), patients were given routine anti-CHF treatment and losartan. However, the patients in control group(n=42) were treated by routine anti-CHF treatment and Benazapril. The heart functional class was evaluated. The content of serum uric acid, electrocardiogram, and Duppler ultransonography were measured before and after treatment in two groups, respectively. Results The variance of heart functional classify, left ventricle diastoic diameter (LVDD), left ventricle ejection fraction (LVEF) and cardiothotic ratio did not show statistical notability between two groups before treament. However, they had been improved sharply compared with base condition in both group after 3 months and 12 months treatment(p﹤0.01) . Although the level of serum uric acid were decreased than base-level in both group after 12 months, but the significant change had shown only in the losartan group (p﹤0.01). Conclusion Losartan is efficacy and safety in the treatment of chronic heart failure with reducing serum uric acid.
    Effects of Propofol on Apoptosis Signaling Pathways in Renal Ischemia/Reperfusion Injury of Rat

    Objective To establish a model of renal ischemic/reperfusion in rats , to observe whether propofol protect kidney from this injury, and deduce a possible mechanism. Methods Ninty-nine male SD rats with right nephrectomy were randomly divided into three groups according to the different treatments to left kidney and rat: Control group (n=26), IR group (n=38), IR+Pro group (n=38). The left kidney was exsected and blood samples were collected 0, 3, 12, 24, 72h postsurgery respectively. Renal morphology were observed with light microscope dyed with HE. Immunohistochemistry was used to detect the expression of apoptosis proteins including Bcl-2、Bax、caspase 3、cytochrome C. Results We observed that renal ischemia-reperfusion induced tissues injury, the severity of injury sequence was: proximal convoluted tubule, distal convoluted tubule, collecting duct, renal corpuscle; According to the results of IHC, the expression of Bax, caspase 3, and cytochrome C in IR group were more than that in Con group, and Bcl-2 level was stable. The protective protein (Bcl-2) level were higher and the proteins (Bax, caspase 3, and cytochrome C ) inducing apoptosis were lower in IR+P group than that of IR group (P<0.05). Conclusions Renal ischemia-reperfusion induced tissues injury and the expression changes of apoptosis proteins; Propofol inhibits the expression of apoptosis proteins such as Bax, caspase 3, cytochrome C, induces the action of Bcl-2. In a word, propofol perhaps inhibit the apoptosis signaling pathways in mitochondrion and cytoplasm.
    Expression of PTEN and MGMT protein in gynecomastia

    Objective To detect the expression of a new tumor suppressor gene PTEN and DNA direct repair enzyme MGMT in gynecomastia. Methods Immunohistochemical SP method was used to detect expression of PTEN and MGMT protein in 68 cases of gynecomastia(experiment group) and 24 cases of mammary gland of normal man tissues(control group). The selected examples were divided into three different age groups and three different histological types. Results The PTEN and MGMT protein were all expressed in nuclear of ductal cellula epithelialis. The expression level of PTEN and MGMT protein in gynecomastia was significantly lower than mammary gland of normal man tissues(P<0.01). There was difference between the expression of PTEN and MGMT protein in different age groups(P<0.05), moreover, the expression level of PTEN protein degraded in middle-aged group, older age-group, and young group by turns;the expression level of MGMT protein degraded in older age-group, middle-aged group, and young group by turns. The expression of PTEN and MGMT protein in three different histological types was different(P<0.05), and the expression level of them degraded in constrictive type, transitional type, and prosperous and intense type by turns. Conclusion This study suggests that the abnormal expression of tumor suppressor gene PTEN and MGMT in gynecomastia probably plays an important role in the development of gynecomastia.

    Objective To identify functional SNPs in the COX-2 promoter and evaluate its effects on risk of endometriosis.Methods The genomic DNA of each subject was extracted by proteinase K digestion followed by saturated hydroxybenzene/ chloroform procedure.Genotypes of COX-2 -1195 promoter of 120 endometriosis cases and 240 controls were determined by polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP).Results The frequencies of GG、AG and AA genotype of COX-2 -1195 promoter were 14.2%、50.0% and 35.8% in 120 patients with endometriosis,24.6%、53.3% and 22.1% in 240 controls respectively, and there was a significant difference between cases and controls. A greater risk of developing endometriosis is observed for A-1195 containing haplotype compared with other haplotypes. It was revealed a 1.63-fold (95%CI=0.87-3.03) and 2.82-fold (95%CI =1.44-5.52) excess risk of developing endometriosis in the individual for -1195A and -1195AA containing haplotype respectively. Conclusion The single nucleotide polymorphism of COX-2-1195G>C promoter may play an important role in genetic basic to endometriosis.

    A phenol-free DNA isolation method from human blood

    Objective To find an ideal method of DNA isolation from blood, especially from clotted blood, to minimize the volume of blood collected for laboratory and clinical tests. Methods DNAs were isolated from antiagglutinating and agglutinating blood samples that were withdrawn from auricular veins of 60 healthy subjects. The DNAs of these samples were obtained by a nonenzymatic, nontoxic procedure optimized by us and determinated by agarose gel electrophoesis and PCR. Results The yields of DNA isolated from clotted blood and antiagglutinating blood were 40.2 ± 8.86 mg /L and 39.1 ± 10.2 mg /L ,and purities were 1.87 ± 0.11 and 1.92± 0.12. The DNAs that we isolated from all samples had high molecular weight and by PCR the dimorphism of Alu alleles of the 8th intron of t-PA was easy to be obtained, so they were complete and reliable. Conclusions The results showed this method is rapid, easy, efficient and nontoxic for isolation of DNA from clotted and fresh blood and it is suit for clinical testing and molecular biology study.
    A model for pancreatic cancer in SD rats and the count of pancreatic myofibroblast

    Objective To establish pancreatic cancer model in Sprague Dawley(SD) rats ,and study the distribution and the counts of myofibroblast(MF) in pancreatic cancer and non-cancerous pancreatic tissues.Methods Directly implanted DMBA into pancreas parenchyma of SD rats and established TSA intervening group and control group. The carcinogenesis of rats executed within 3-5 months were inspected by HE stain and microscope for the study of pathomorphological changes.And myofibroblast(MF) was stained by Heidenains method for the counts under microscopy of high fields. Results (1)The prevalence of pancreatic cancer in experimental group was 48.7%(18/37), including 17 pancreatic ductal adenocarcinoma and 1 fibrosarcoma. The prevalence of pancreatic cancer in intervering group was 33.3%(12/36), including 11 pancreatic ductal adenocarcinoma and 1 fibrosarcoma.The prevalence of pancreratic carcinoma was significantly in experimental group and in intervening group than that in control group (0/10,P<0.05 or P<0.01).The maximal diameters of tumor mass of experimental group was higher than that of intervering group (P<0.05).(2) The counts of MF were significantly higher in pancreatic cancer of experimental group and intervering group than those in non-cancerous pancreatic tissues of experimental group and intervering group (P<0.01). The counts of MF were significantly higher in moderately or severely atypical ductal hyperplastic tissues than those in normal tissues or mild atypical ductal hyperplastic tissues in non-cancerous pancreatic tissues(P<0.05). The counts of MF were significantly higher in non-cancerous or cancerous pancreatic tissues of experimental or intervering group than those in control group (P<0.05 or P<0.01). Conclusions Pancreatic MF might have an important effect on the carcinogenesis and progression of pancreatic cancer.

    Objective To study serum proteomic fingerprints of colorectal cancer during onset and progression and to screen tumor markers related to prognosis. Methods Serum from colorectal cancer patients, non-cancer patients, and healthy control were profiled using WCX ProteinChip or magnetic beads and analyzed by mass spectrometry. Results Seven protein peaks were found related to colorectal cancer. Several peaks were closely related to lymph node metastasis, distal organ metastasis and decreased after surgery. The diagnostic model composed of 3398.3、5477.1、8453.9 u can detect CEA negative colorectal cancer in 100%. Conclusion Protein fingerprinting technology (PFT) in conjunction with bioinformatics can significantly identify novel biomarkers in the serum of colorectal cancer patients with potential values for prognostic evaluation, detection of CEA negative colorectal cancer and changing its progression.

    Objective To compare the effects of equivalent dose of sufentanil and fentanyl on wake-up test and recovery profile in scoliosis surgery. Methods Forty ASAⅠ-Ⅱadolescents undergoing scoliosis surgery were randomly divided into fentanyl group (group F,n=20) and sufentanil group(group S,n=20). Anesthesia was maintained with low-flow(1L/min) inhalation of isoflurane and nitrous oxide (O2:N2O=1:1) in both groups. Intermittent i.v. boluses (1~1.5μg/kg) of fentanyl was used for analgesia in group F, and total dosage was no more than 5μg/kg when the wake-up test was started. Continuous infusion of sufentanil (0.1~0.2μg?kg-1?h-1) was maintained in group S, and total dosage was less than 1μg/kg when the wake-up test was started. The wake-up test time, postoperative recovery time, end-tidal isoflurane concentration(ETiso) and other hemodynamic variables during operation were recorded. Results There were no significant differences in the wake-up test time, postoperative recovery time, bucking and/or restlessness during the wake-up test, PCA morphine dosage and PONV between the two groups. Conclusion Properly administration of sufentanil does not prolong the intraoperative wake-up test time so that it can be used safely in scoliosis surgery.

    Langerhans' cell histiocytosis is a kind of uncommon disease , involvement of the thyroid by langerhans' cell histiocytosis is rare. 2 cases of Langerhans cell histiocytosis of the thyroid confirmed by pathology were reported in the paper. The main clinical represent was the enlargement of the thyroids. One patient had hypothyroidism. Meanwhile, the lungs and pituitaries of the two patients were affected. Langerhans' cell histiocytosis of the thyroid has no typical symptoms and specific laboratory examination, and can be clinically confused with chronic lymphocytic thyroiditis, benign and malignant thyroid tumors. Thyroid fine needle aspiration cytology may be useful in making the diagnosis. The effective treatments include systemic chemotherapy, local radiation therapy and surgical excision. The chemotherapy is still the main approach. Most patients can relieve partially after receiving the systemic chemotherapy.
