Effects of S-adenosylhomocysteine on proliferation and apoptosis of rat SV40-transformed aortic rat endothelial cell

Abstract: Objective To study the effect of AdoHcy on proliferation and apoptosis of SVAREC and the content of VCAM-1 in the supernatant of SVAREC culture.Methods After SVAREC were treated with different concentrations of 3-deazaadenosine (DZA) for 24, 48 and 72h, the cellular AdoHcy level and the homocysteine (Hcy) were measured with different methods The cell growth and proliferation were determined by WST-1 assay. The percentage was analyzed with TUNEL methods. The content of VCAM-1 in the supernatant of SVAREC culture was analyzed with ELISA methods Results After SVAREC were treated with different concentrations of DZA, the content of Hcy decreased while the intracellular AdoHcy level increased.Compared with the control group, AdoHcy could inhibit the proliferation and induce apoptosis of SVAREC in a dose-dependent way and induce secretion of VCAM-1. Conclusions AdoHcy could inhibit the proliferation and induce apoptosis of SVAREC, induce secretion of VCAM-1.which may be one of mechanisms responsible for injury of aorta endothelial cell lead to atherosclerosis.
