Abstract: Objective To improve methods of primary culturing rats myocadial microvascular endothelial cells in vitro, and observing the growth feature. Methods Enzyme digestion cultivation of cadiac tissue was used to obtain the primary cells. In the subculturing, the cell cultures were purified by different speeds of attachment and sub-cell cloning method. The purified cells were identified as endothelial cells by factor Ⅷ and CD31 related antigen ,the two markers for endothelial cells, and observing the growth feature by inverted phase contrast microscope. Results The MMVECs were cultured and purified successfully which can be used in the related researches. Conclusion The protocol of culturing MMVECs was succeeded, and it is reliable.
