Lnc-SLC2A12-10:1 silencing inhibits the proliferation, migration and invasion of gastric cancer cell line AGS

Abstract: Objective To investigate the effect of long non-coding RNA(lncRNA) lnc-SLC2A12-10:1 silencing on the proliferation, migration and invasion of gastric cancer cell line AGS. Methods Gene interference technology was used to construct the lnc-SLC2A12-10:1 silenced gastric cancer cell strain AGS, and RT-qPCR was used to verify the interference efficiency by lnc-SLC2A12-10:1 silencing. The proliferation of AGS cell was examined by CCK-8 method. The cell migration and invasion were evaluated by wound healing and Transwell assay. Bioinformatics prediction software was applied to predict lnc-SLC2A12-10:1 targeting miRNA and to verify them with RT-qPCR. Results Lnc-SLC2A12-10:1 shRNA effectively silenced expression of lnc-SLC2A12-10:1, and lnc-SLC2A12-10:1 silencing inhibited the proliferation of gastric cancer cells (P<0.05). Lnc-SLC2A12-10:1 silencing also inhibited the migration and invasion of AGS cells (P<0.05). It had been demonstrated that lnc-SLC2A12-10:1 targeting miRNAs might be hsa-miR-105-5p and hsa-miR-150-3p. Conclusions Lnc-SLC2A12-10:1 silencing can significantly inhibit the proliferation, migration and invasion of AGS cells.
