Up-regulation expression of miR-195-5p inhibits the migration and invasion of lung cancer cell line A549

Abstract: Objective To investigate the effect and mechanism of miR-195-5p on the migration and invasion of A549 cells in vitro. Methods The expression of miR-195-5p in lung cancer tissues, paracancerous tissues and lung cancer cell lines were analyzed by quantitative real-time PCR. Moreover, miR-195-5p mimics were transfected into A549 cells by liposome-mediated method. The cell migration and invasion were examined by Transwell assay. Western blot analysis was used to detect the effect of miR-195-5p on the protein level expression of epithelial-mesenchymal transition makers and ZEB1. Results Compared with paracancerous tissues and normal lung epithelial cells, the expressions of miR-195-5p were lower in lung cancer tissues and lung cancer cell line(P <0.001). The over-expression of miR-195-5p inhibited the migration and invasion of A549 cells(P <0.001), increased the protein expression of E-cadherin and reduced expression of N-cadherin、vimentin and ZEB1(P <0.01). Conclusions miR-195-5p inhibits the migration and invasion of lung cancer cells by reducing the expression of ZEB1.
