Effcects of Interleukin-1β on the expression of VEGF In A549 cells lines

Abstract: Abstract: Objective To investigate the effect of interleukin-1β(IL-1β) on the expression of VEGF in A549 cells lines. Methods: A549 cells were incubated with IL-1βin the presence or absence of Celecoxib. After indicated incubation time, cells were harvested to detect VEGFmRNA by reverse transcription-polymerase chain rection (RT-PCR), and the medium was collected to determine the concentration of VEGF protein by enzyme-linked immunosorbent assay (ELISA).Results: After treated with IL-1β, the expression of VEGF mRNA and protein in A549 cells were significantly higher than that in the control group(P<0.05); IL-1β upregulate the expression of VEGF mRNA, protein in a time-dependent and dose-dependent manner in A549 cells, after incubation 24 hours, VEGF expression were up to high. After Celecoxib intervention, VEGF expression were decrease than control group(IL-1βinduced), the difference was significance(P<0.05); with the concentration of Celecoxib step up, VEGF expression were decrease in dose-dependent manner, and the difference within different group was significance(P<0.05. Conclusions: These results suggest that IL-1β upregulate VEGF expression in A549 cells lines,the mechanism may be at transcriptional level; Celecoxib down-regulated IL-1β induced VEGF expression in A549 cells lines.
