In vitro differentiation of rat bone marrow mesenchymal stem cells into dopamine-like nerve cells

Abstract: To explore the differentiation of rat bone marrow mesenchymal stem cells into dopamine nerve cells. The mesenchymal stem cells were isolated and purified by cell culture base on growth characteristics of bone marrow cells. When passed third generation, they were induced to differentiate into dopamine nerve-like cells by bFGF, Vitc and EFGF. Dopamine-associated protein and genes in the treated cells was examined by immumofluorescence method and RT-PCR. Dopamine in the supernatant and cytoplasm from culture system was determined by ELISA kit. The results show that Tyrosine hydroxylase, dopamine transporter and nerve neucleprotein and Nestin,Nurr-1 genes were found. And the dopamine exists in in the supernatant and cytoplasm from inducing culture system. It is suggested rat bone marrow mesenchymal stem cells have the capacity of differente into dopamine nerve cells.
